蜘蛛香单帖环烯醚类成分及其抗乳腺癌活性

Iridoids from Valeriana jatamansi and their anti breast cancer activity

目的研究蜘蛛香Valeriana jatamansi Jones.单帖环烯醚类成分及其抗乳腺癌活性。方法蜘蛛香95%乙醇提取物采用正相和反相硅胶、中压柱、制备型HPLC、反相HPLC进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。MTT法测定其抗乳腺癌活性。结果从中分离得到9个化合物,分别鉴定为valtrate(1)、desoxidodidrovaltrate(2)、1-homovaltratum(3)、nardostachin monoacetate(4)、11-ethoxyviburtinal(5)、valeriandoid B(6)、volvaltrate B(7)、baldrinal(8)、isodidrovaltratum(9)。化合物1~4、6对人源三阴性乳腺癌细胞MDA-MB-231有一定的抑制作用,IC 50分别为(19.02±0.23)、(23.49±0.19)、(24.76±0.37)、(21.03±0.09)、(26.35±0.24)μmol/L;化合物6、8对人源乳腺癌细胞MCF-7有一定的抑制作用,IC 50分别为(35.4±0.42)、(37.39±0.16)μmol/L。结论化合物4为首次从该属植物中分离得到,化合物1为首次从该植物中分离得到。化合物1~4、6、8对人乳腺癌细胞有较显著抑制作用。

AIM To study iridoids from Valeriana jatamansi Jones.and their anti breast cancer activity.METHODS The 95%ethanol extract from V.jatamansi was isolated and purified by normal phase and reversed phase silica,medium pressure column,preparative HPLC and reversed-phase HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.The anti breast cancer activities were determined by MTT mothod.RESULTS Nine compounds were isolated and identified as valtrate(1)、desoxidodidrovaltrate(2)、1-homovaltratum(3)、nardostachin monoacetate(4)、11-ethoxyviburtinal(5)、valeriandoid B(6)、volvaltrate B(7)、baldrinal(8)、isodidrovaltratum(9).Compound 1-4、6 had a certain inhibitory effect on MDA-MB-231 cells with the IC_(50) value of(19.02±0.23),(23.49±0.19),(24.76±0.37),(21.03±0.09),(26.35±0.24)μmol/L,respectively.And compound 6,8 had a certain inhibitory effect on MCF-7 cells with the IC_(50) value of(35.4±0.42),(37.39±0.16)μmol/L,respectively.CONCLUSION Compound 4 is isolated from genus Valeriana for the first time,and compounds 1 is isolated from this plant for the first time.Compounds 1-4,6 and 8 have significant inhibitory effect on human breast cancer cells.
