摘要 为明确人工无机窖泥发酵过程中的梭菌群落多样性,利用聚合酶链式反应-变性梯度凝胶电泳技术对窖泥样品中的梭菌群落结构及变化规律进行研究。结果表明,窖泥样品中含有丰富的梭菌资源,主要有梭菌属(Clostridium)、丁酸弧菌属(Butyrivibrio)、罗氏菌属(Roseburia)、醋弧菌属(Acetivibrio)、Alkalibaculum、Gracilibacter、Anaerocolumna、Sporanaerobacter、Sedimentibacter、互营球菌属(Syntrophococcus)、厌氧醋菌属(Acetoanaerobium)、真杆菌属(Eubacterium)以及巨型球菌属(Megasphaera)。经过发酵,大部分梭菌都能在窖泥中生长。梭菌属、丁酸梭菌、真杆菌是发酵过程中的优势菌,对窖泥的特性起重要作用。人工窖泥发酵过程中多样性指数呈下降后螺旋上升的变化过程,菌群有一定的亲缘性,菌落结构变化较小。 In order to ascertain the diversity of Clostridia species during the fermentation process of artificial inorganic pit mud,polymerase chain reaction-denaturing gradient gel electrophoresis(PCR-DGGE)was adopted to study the community structure and change rules of Clostridia in the pit mud samples.The results show that there are abundant Clostridia resources in the pit mud,and the main genera are Clostridium,Butyrivibrio,Roseburia,Acetivibrio,Alkalibaculum,Gracilibacter,Anaerocolumna,Sporanaerobacter,Sedimentibacter,Syntrophococcus,Acetoanaerobium,Eubacterium,and Megasphaera.After fermentation,most Clostridia species can grow in the pit mud,and the dominant genera are Clostridium,Butyrivibrio and Eubacterium,which contribute greatly to the properties of the pit mud.In the fermentation process of the artificial pit mud,the diversity index decreases firstly and then spirals upward,the microbial communities are related,and the change of the community structure is small.
