摘要
目的探讨通过核因子-κB(NF-κB)寡脱氧核苷酸诱骗剂(ODN Decoy)构建的耐受性树突状细胞(DC)对胶原诱导性关节炎(CIA)大鼠血清中IL-4、TGF-β、IFN-γ和IL-17表达水平的影响。方法以24只雌性SD大鼠为实验动物,随机将其分为空白对照组、CIA模型组、BⅡC-Decoy DC实验组和BⅡC-Decoy DC对照组,每组6只。CIA模型组在大鼠左足注射牛Ⅱ型胶原(BⅡC),空白对照组注射生理盐水,BⅡC-Decoy DC实验组在CIA模型组的基础上在初次免疫第20天尾静脉输注BⅡC-Decoy DC,BⅡC-Decoy DC对照组在初次免疫后第20天注射BⅡC-Decoy DC,其余处理同空白对照组。从建模后观察各组大鼠关节炎性症状和关节滑膜组织病理形态,以ELISA法检测各组大鼠血清IL-4、TGF-β、IFN-γ和IL-17水平。结果与CIA模型组相比,BⅡC-Decoy DC实验组大鼠关节炎症状、滑膜组织病理表现减轻,并且血清IL-4、TGF-β水平显著升高,IFN-γ、IL-17水平显著降低(P<0.05)。结论耐受性DC可通过纠正CIA大鼠血清IFN-γ/IL-4、IL-17/TGF-β失衡来减轻关节炎症等症状。
Objective To explore the effects of tolerant dendritic cells(DC)constructed by nuclear factor-κB(NF-κB)oligodeoxynucleotide Decoy(ODN Decoy)on serum IL-4,TGF-β,IFN-γ and IL-17 expression levels in collagen-induced arthritis(CIA)rats.Methods Taking 24 female SD rats as experimental animals,the animals were randomly divided into blank control group,CIA model group,BⅡC-Decoy DC experimental group and BⅡC-Decoy DC control group,with 6 rats in each group.The CIA model group was injected with bovine typeⅡcollagen(BⅡC)on the left foot,the blank control group was injected with saline,the BⅡC-Decoy DC experimental group was injected with BⅡC-Decoy DC through caudal vein on the 20th day after the initial immunization based on the CIA model group,the BⅡC-Decoy DC control group was injected with BⅡC-Decoy DC on the 20th day after the initial immunization,and the other treatments were the same as the blank control group.After modeling,the arthritis symptoms and pathological morphology of synovium tissue of joint of rats were observed,and the serum IL-4,TGF-β,IFN-γ and IL-17 levels were detected by ELISA.Results Compared with the CIA model group,the arthritis symptoms and pathological manifestations of synovium tissue of joint of rats in the BⅡC-Decoy DC experimental group reduced,the serum IL-4 and TGF-β levels significantly increased,and the IFN-γ and IL-17 levels significantly reduced(P<0.05).Conclusion Tolerant DC can reduce the symptoms of arthritis by correcting the imbalance of serum IFN-γ/IL-4 and IL-17/TGF-β in the CIA rats.
