摘要
目的探讨miR-552-3p对非小细胞肺癌细胞增殖、迁移及侵袭的影响及其调控Wnt信号通路的机制。方法分别将mi-NC、miR-552-3p转染至NSCLC A549细胞内,根据转染物的不同将其分为NC组和mimic组,设置正常NSCLC A549细胞为对照组。转染6、12、24、48、72 h后,采用CCK-8法检测三组细胞的增殖能力;转染48 h后,采用Transwell小室实验检测三组细胞的迁移、侵袭能力,采用Western blot检测三组细胞Wnt、β-catenin、c-Myc蛋白的相对表达量。结果mimic组转染12、24、48、72 h后NSCLC A549细胞增殖率均低于对照组和NC组,差异具有统计学意义(P<0.05)。mimic组的相对细胞迁移率、细胞侵袭率明显低于对照组,差异具有统计学意义(P<0.05)。mimic组的Wnt、β-catenin、c-Myc蛋白相对表达量明显低于对照组和NC组,差异具有统计学意义(P<0.05)。结论miR-552-3p能够抑制NSCLC A549细胞增殖、迁移及侵袭,其可能通过靶向调控Wnt信号通路发挥此作用。
Objective To investigate the effects of miR-552-3p on proliferation,migration and invasion of non-small cell lung cancer(NSCLC)cells and its mechanism of regulating Wnt signaling pathway.Methods The mi-NC and miR-552-3p were transfected into NSCLC A549 cells respectively.According to the different transfectants,NSCLC A549 cells were divided into NC group and mimic group,and normal NSCLC A549 cells were set as control group.After transfection for 6,12,24,48 and 72 h,the proliferation ability of cells in the three groups was detected by CCK-8 method;after transfection for 48 h,the migration and invasion ability of cells in the three groups were detected by Transwell chamber experiment,and the relative expressions of Wnt,β-catenin and c-Myc proteins of the three groups were detected by Western blot.Results After transfection for 12,24,48 and 72 h,the proliferation rates of NSCLC A549 cells in the mimic group were lower than those in the control group and the NC group,and the differences were statistically significant(P<0.05).The relative cell migration rate and cell invasion rate in the mimic group were significantly lower than those in the control group,and the differences were statistically significant(P<0.05).The relative expressions of Wnt,β-catenin and c-Myc proteins in the mimic group were significantly lower than those in the control group and the NC group,and the differences were statistically significant(P<0.05).Conclusion The miR-552-3p can inhibit the proliferation,migration and invasion of NSCLC A549 cells,which may play these roles through targeted regulation of Wnt signaling pathway.
