摘要 该文研究表面活性剂协同双频超声波提取香菇多糖工艺及其抗氧化活性。以香菇多糖(lentinan,LNT)提取率为指标,采用单因素试验和正交试验,确定最优提取工艺参数。结果表明,最优工艺参数为料液比1∶40(g/mL)、表面活性剂用量2.5%、超声频率(25+40)kHz、超声时间25 min,在此条件下,LNT提取率为14.16%。抗氧化试验结果表明,在一定的质量浓度范围内,LNT对DPPH自由基和羟基自由基的清除率随着浓度的增加而升高,当LNT质量浓度为25 mg/mL时,其对DPPH自由基和羟基自由基的清除率分别为78.51%和82.28%,说明LNT具有较强的抗氧化活性。表面活性剂协同双频超声波辅助提取香菇多糖的方法稳定、可行。 The extraction technology and antioxidant activity of lentinan by surfactant ultrasonic-assisted dual-frequency were studied.Considering the extraction rate of lentinan(LNT)as the index,the single factor and orthogonal tests were used to determine the optimal extraction process parameters.The optimum process parameters obtained were as follows:solid-liquid ratio 1∶40(g/mL);surfactant dosage 2.5%;ultrasonic frequency(25+40)kHz;and ultrasonic time 25 min.Under these conditions,the extraction rate of LNT was 14.16%.The antioxidant test results showed that in a certain concentration range,the scavenging rates of LNT on 2,2-diphenyl-1-picrylhydrazyl(DPPH)and hydroxyl radicals increased with the increase in concentration.When the mass concentration of LNT was 25 mg/mL,the scavenging rates of LNT on DPPH and hydroxyl radicals were 78.51%and 82.28%,respectively,indicating that LNT has strong antioxidant activity.The method of extracting lentinan by surfactant ultrasonic-assisted dual-frequency were stable and feasible.
