马齿苋提取物对急性湿疹模型大鼠皮肤屏障功能及炎症因子和免疫因子表达的影响 被引量:13

摘要 目的研究马齿苋提取物对急性湿疹模型大鼠皮肤屏障功能及炎症因子和免疫因子表达的影响。方法将50只无特定病原体(SPF)级SD大鼠按照随机数字表法分为对照组、模型组和高、中、低剂量马齿苋组,每组10只。模型组和3组马齿苋组采用2,4-二硝基氯苯(DNCB)建立急性湿疹模型,造模成功后,对照组和模型组大鼠给予1 mL无菌蒸馏水,高、中、低剂量马齿苋组大鼠分别给予相当于生药含量为2、1、0.5 g/mL马齿苋提取液各1 mL,每天给药2次,共治疗9 d。分别于D3和D9时采用多功能皮肤测试仪检测3组大鼠的经皮水分丢失量(TEWL)的情况;免疫组织化学测定皮肤组织中肿瘤坏死因子-α(TNF-α)和白细胞介素-4(IL-4)的表达;酶联免疫吸附试验(ELISA)法检测血清中干扰素-γ(IFN-γ)和免疫球蛋白E(IgE)的表达水平。结果3组马齿苋组的TEWL在D3时与模型组比较,差异无统计学意义(P>0.05),而在D9时则显著低于模型组,差异有统计学意义(P<0.05)。与模型组相比,3组马齿苋组在D3时的大鼠皮肤组织中TNF-α和IL-4的表达量差异无统计学意义(P>0.05),而在D9时3组马齿苋组的TNF-α和IL-4的表达量均明显低于模型组,差异均有统计学意义(P<0.05)。与模型组相比,3组马齿苋组在D3时血清中IFN-γ和IgE的表达水平差异无统计学意义(P>0.05),而在D9时3组马齿苋组的IFN-γ的表达水平均高于模型组、IgE的表达水平均低于模型组,差异均有统计学意义(P<0.05)。结论马齿苋提取物对急性湿疹疗效显著,其作用机制可能与抑制TNF-α、IL-4和IgE的表达、提高IFN-γ的表达、并恢复皮肤屏障功能有关。 Objective To investigate the effect of purslane extract on skin barrier and expressions of inflammatory and immune factors in rats with acute eczema.Methods A total of 50 specific pathogen free(SPF)grade SD rats were divided into control group,model group and high-,middle-,low-dosage purslane group according to the random number table method,with 10 rats in each group,the acute eczema model was established by dinitrochlorobenzene(DNCB).The control group and model group were treated with 1mL distilled water,three purslane groups were treated with 2,1,0.5 g/mL purslane extract 1mL respectively,twice a day for 9 days.The transepidermal water loss(TEWL)was detected by multifunctional skin tester at D3 and D9;the expression of tumor necrosis factor-α(TNF-α)and interleukin-4(IL-4)in skin tissue were detected by immunohistochemistry;the expression level of serum interferon-γ(IFN-γ)and immunoglobulin E(IgE)were detected by enzyme-linked immunosorbent assay(ELISA).Results There was no significant difference of TEWL between three purslane groups and model group at D3(P>0.05),while the TEWL of three purslane groups were lower than model group at D9,the difference was statistically significant(P<0.05).There was no significant difference in the expression of TNF-αand IL-4 in the rat skin tissue in three purslane groups and model group at D3(P>0.05).The expression levels of TNF-αand IL-4 in the three portulaca groups were significantly lower than the model group at D9,and the differences were statistically significant(P<0.05).There was no significant difference in the expression levels of serum IFN-γand IgE in the three groups of portulaca and the model group at D3(P>0.05).The expression level of IFN-γin the three portulaca groups was higher than that of the model group at D9,and the expression level of IgE was lower than that of the model group at D9,the differences were statistically significant(P<0.05).Conclusion Purslane extract had siginificant efficacy in the treatment of acute eczema,which probably related to inhibition of TNF-α,IL-4 and IgE,increase of IFN-γ,and restore the skin barrier functions.
