白杨素对舌鳞状细胞癌CAL-27细胞活力、形态及凋亡的影响

Effects of chrysin on viability,morphology and apoptosis of CAL-27 cells in tongue squamous cell carcinoma

目的探讨白杨素对舌鳞状细胞癌CAL-27细胞活力、形态、凋亡的影响。方法体外培养舌鳞状癌细胞CAL-27,空白对照组不给予任何药物,DMSO组以0.1%DMSO试剂处理,白杨素组给予不同浓度白杨素(1.0、2.5、5.0、10.0、15.0、20.0μmol/L)处理。MMT法检测细胞活力,采用倒置显微镜观察细胞形态变化,TUNEL法检测细胞凋亡,Western blot印迹法检测凋亡蛋白(Bcl-2、Caspase-3)的表达情况。结果CAL-27细胞经不同浓度白杨素处理24 h后,其细胞增殖活性被抑制;白杨素对CAL-27细胞形态存在显著影响,且凋亡小体数量增加;白杨素可促进CAL-27细胞凋亡(P<0.05);白杨素可诱导Bcl-2表达显著下降及Caspase-3活性表达显著上升(P<0.05)。结论白杨素可以抑制舌鳞状细胞癌CAL-27细胞增长,降低细胞活力,调控细胞凋亡蛋白Bcl-2、Caspase-3活性的表达,促进细胞凋亡。

Objective To investigate the effects of chrysin on the viability,morphology and apoptosis of CAL-27 cells in tongue squamous cell carcinoma.Methods The tongue squamous carcinoma cell CAL-27 were cultured in vitro.The blank control group was not given any drugs,the DMSO group was treated with 0.1%DMSO reagent,and the chrysin group was treated with different concentrations of chrysin(1.0,2.5,5.0,10.0,15.0,20.0μmol/L).The MMT method was used to detect the cell viability.The inverted microscope was used to observe the morphological changes of cells.The TUNEL method was used to detect the apoptosis of cells.And Western blot method was used to detect the expression of apoptotic proteins(Bcl-2,Caspase-3).Results The proliferation activity of CAL-27 cells was inhibited after treatment with different concentrations of chrysin for 24 hours.Chrysin had a significant effect on the morphology of CAL-27 cells and the number of apoptotic bodies was increased.Chrysin could promote the apoptosis of CAL-27 cells(P<0.05).Chrysin could induce a significant decrease in the expression of Bcl-2 and a significant increase in the expression of Caspase-3 activity(P<0.05).Conclusion Chrysin can inhibit the growth of CAL-27 cells in tongue squamous cell carcinoma,reduce the cell viability,regulate the expression of apoptotic proteins activities of Bcl-2 and Caspase-3,and promote the apoptosis.
