摘要
目的研究黄体生成素/人绒毛膜促性腺激素受体(LHCGR)、卵泡刺激素受体(FSHR)、Yes相关蛋白1(YAP1)DNA多态性与多囊卵巢综合征(PCOS)发病的相关性。方法选取2019年10月-2021年2月新疆医科大学第一附属医院生殖医学中心PCOS患者59例为PCOS组,以同期健康体检者59例为对照组。采取聚合酶链反应及焦磷酸测序法测定两组LHCGR、FSHR、YAP1DNA多态性,对比两组一般资料及上述基因多态性位点基因型分布频率,采用Logistic回归方程分析PCOS发病的相关因素。结果与对照组比较,PCOS组LH、T指标升高,FSH指标降低,差异有统计学意义(P<0.05);PCOS组rs13405728位点基因型TT、TC、CC频率分别为69.49%、28.81%、1.69%;rs1394205位点基因型AA、AG、GG频率分别为59.32%、22.03%、18.64%;rs11225161基因型AA、AG、GG频率分别为18.64%、59.32%、22.03%;对照组rs13405728位点基因型TT、TC、CC频率分别为50.85%、27.12%、22.03%;rs1394205位点基因型AA、AG、GG频率分别为55.93%、20.34%、23.73%;rs11225161基因型AA、AG、GG频率分别为44.07%、47.46%、8.47%。两组rs13405728位点基因型频率、rs11225161基因型分布频率比较,差异有统计学意义(P<0.05)。结果显示,rs13405728位点基因型、rs11225161位点基因型多态性均与PCOS发生有关(P<0.05)。结论LHCGR、YAP1DNA是PCOS的易感基因,其中rs13405728位点T/C多态、rs11225161位点A/G多态与PCOS易感性存在关联性。
Objective To study the relationship between luteinizing hormone/human chorionic gonadotropin receptor(LHCGR),follicle stimulating hormone receptor(FSHR),Yes-related protein 1(YAP1)DNA polymorphisms and poly⁃cystic ovary syndrome(PCOS).Methods From October 2019 to February 2021,59 patients with PCOS in the hospital were selected as PCOS group,and 59 patients with healthy physical examination during the same period were selected as control group.The two groups of LHCGR,FSHR,YAP1 DNA polymorphisms were measured by polymerase chain reaction and pyrosequencing methods.The general data of the two groups and the genotype distribution frequency of the above gene polymorphism loci were compared.The logistic regression equation was used to analyze the related fac⁃tors of PCOS.Results Compared with the control group,luteinizing hormone and testosterone indexes in PCOS group increased and FSH indexes decreased(P<0.05);the frequencies of genotypes TT,TC,and CC at rs13405728 in the PCOS group were 69.49%,28.81%,and 1.69%respectively;the frequencies of genotypes AA,AG,and GG at rs1394205 were 59.32%,22.03%,and 18.64%respectively;the frequencies of genotypes AA,AG,GG at rs11225161 were 18.64%,59.32%,and 22.03%respectively;the frequencies of genotypes TT,TC,and CC at rs13405728 in the control group were 50.85%,27.12%,and 22.03%respectively;the frequencies of genotypes AA,AG,and GG at rs1394205 were 55.93%,20.34%,and 23.73%respectively;the frequencies of genotypes AA,AG,GG at rs11225161 were 44.07%,47.46%,and 8.47%respectively.There was significant difference in genotype distribution frequency of rs13405728 and rs11225161 between the two groups(P<0.05).The rs13405728 genotype and the rs11225161 geno⁃type polymorphisms were all associated with PCOS occurrence(P<0.05).Conclusion LHCGR and YAP1 DNA are the susceptibility genes of PCOS.Among them,the T/C polymorphism at rs13405728 and the A/G polymorphism at rs11225161 are associated with PCOS susceptibility.
