摘要
肝纤维化是慢性肝病进程中肝脏反复损伤和修复的结果,是以细胞外基质过度累积为主要特征的病理过程。肝星状细胞异常活化是肝纤维化的细胞学基础,其活化受多种因素影响,其中,缺氧诱导因子-1α(HIF-1α)发挥了重要作用。HIF-1α作为机体适应缺氧应激的关键细胞因子,对肝星状细胞的活化、增殖和促细胞外基质形成、促血管新生能力的改变有重要意义。中医药可通过调控HIF-1α的表达以及改善肝脏乏氧微环境减轻肝纤维化的程度,因此HIF-1α有望成为中医药防治肝纤维化的新靶点。本文就HIF-1α的概述、HIF-1α调控肝星状细胞活化、自噬、凋亡等功能以及中医药调控HIF-1α的表达防治肝纤维化等方面作一综述。
Liver fibrosis is a pathological process characterized by excessive deposition and accumulation of extracellular matrix(ECM), which is the result of repeated liver damage and repair during a wide range of chronic liver injures.Hepatic stellate cells(HSCs), which abnormal activation is affected by many cytokines and signal pathways, result in liver fibrosis. As a key cytokine adapted to hypoxic stress, hypoxia-inducible factor-1 α(HIF-1α) plays an important role in the activation and proliferation of HSCs. Meanwhile, the capability of ECM formation and angiogenesis is improved. In addition, traditional Chinese medical treatment towards the HIF-1α associated signaling pathway could relieve the process of liver fibrosis, which suggests it may be a potential molecular target for liver fibrosis treatment. This article reviewed the influence of HIF-1α on HSCs comprehensively, including the structure and function of HIF-1α,activation, autophagy and apoptosis regulated by HIF-1α, and traditional Chinese medicine treatment for HIF-1α.
