摘要
建立一种高效液相色谱法测定婴幼儿乳粉中乳铁蛋白含量的方法,用磷酸盐缓冲液提取,磷酸调pH,除酪蛋白,高效液相色谱法测定,外标法定量。以HiTrapTM Heparin HP(1 mL)肝素亲和柱为色谱柱,磷酸盐缓冲液为流动相,梯度洗脱,检测波长为280 nm。结果表明,平均加标回收率(n=6)在89.9%~101.0%,相对标准偏差(RSD)在1.61%~4.00%,定性检测限(LOD)是0.1 mg/100 g,定量检测限(LOQ)是0.3 mg/100 g。该方法前处理简单,结果准确,精密度高,重现性好,可应用于婴幼儿乳粉中乳铁蛋白的定量分析。
Objective to establish a method for the determination of lactoferrin in infant milk powder by high performance liquid chromatography(HPLC).Hitraptm heparin HP(1 mL)heparin affinity column was used as chromatographic column,phosphate buffer as mobile phase,gradient elution,detection wavelength was 280 nm.The results showed that the average recoveries(n=6)ranged from 89.9%to 101.0%,the relative standard deviations(RSD)ranged from 1.61%to 4.00%,the limits of qualitative detection(LOD)and quantitative detection(LOQ)were 0.1 mg/100 g and 0.3 mg/100 g respectively.The method has the advantages of simple pretreatment,accurate results,high precision and good reproducibility.It was applied to the quantitative analysis of lactoferrin in infant milk powder.
