摘要
目的探讨气阴两虚型非小细胞肺癌肠道菌群、代谢组学和蛋白组学的基本特征。方法选择气阴两虚型非小细胞肺癌患者13例作为患者组,选择健康志愿者15名作为健康对照组,收集两组粪便标本,用16S rDNA技术检测肠道微生态。采集两组血清标本,通过液相色谱-质谱联用技术比较两组代谢组学和蛋白质组学差异。最后通过WGCNA软件对多组学结果进行联合分析。结果两组间存在6种显著差异菌,20种差异蛋白(上调5个,下调15个),11种差异代谢物(负离子模式下2个,正离子模式下9个),差异菌群、差异蛋白与差异代谢物间存在相关性,联合分析发现差异物质与癌症通路相关。结论气阴两虚型非小细胞肺癌具有特异的肠道菌群、蛋白质和代谢物,与肿瘤的发生发展密切相关。
Objective To explore the characteristics of intestinal flora,metabolomics,and proteomics in non-small cell lung cancer(NSCLC)with deficiency in both Qi and Yin.Methods This study recruited 13 NSCLC patients with deficiency in both Qi and Yin and 15 healthy volunteers.Fecal samples of these two groups of patients were collected and intestinal microflora was analyzed by sequencing the 16S rDNA.Serum samples were also collected from two groups of patients and used for the analysis of metabolomics and proteomics using liquid chromatography-mass spectrometry.After that,a joint analysis of multi-omics was carried out using the weighted gene co-expression network analysis(WGCNA).Results The data revealed six significantly different kinds of bacteria,20 differentially expressed proteins(5 upregulated and 15 downregulated),and 11 different kinds of metabolites(2 in negative ion mode and 9 in positive ion mode)between two groups.There was an association among differential bacteria,proteins and metabolites in these two groups of samples.WGCNA data showed that such differences in bacteria,proteins and metabolites in these two groups of samples were associated with"the cancer pathway".Conclusion NSCLC patients with deficiency in both Qi and Yin have specific intestinal flora,protein expression,and metabolites,which were closely associated with NSCLC development.
