摘要
目的研究白杨素衍生物dFMAChR对裸鼠人乳腺癌MCF-7细胞原位移植瘤生长的影响及其可能机制。方法裸鼠原位皮下注射人乳腺癌MCF-7细胞构建动物模型,给予梯度浓度dFMAChR后测定移植瘤大小和最终重量;瘤组织切片后行HE染色和免疫组化,分别考察肿瘤组织病理变化、VEGF蛋白的表达情况。结果20、40和80 mg·kg^(-1)的dFMAChR可抑制肿瘤生长,抑瘤率分别为40.3%、55.4%和75.0%。HE和免疫组化结果表明dFMAChR可通过下调瘤组织中VEGF蛋白的表达,杀伤肿瘤细胞。结论dFMAChR可通过下调VEGF蛋白剂量依赖性抑制人乳腺癌原位移植瘤的生长。
OBJECTIVE To evaluate the effect of chrysin derivative(dFMAChR)on the growth of human breast cancer xenografts in nude mice and its possible mechanism.METHODS Balb/c nude mice bearing MCF-7 xenografts were established and the size and final weight of the transplanted tumor were measured after treatment of gradient concentrations of dFMAChR.HE staining and immunohistochemistry were performed to investigate the pathology of tumor tissues and the expression of VEGF protein.RESULTS The tumor inhibition rates of 20,40 and 80 mg·kg^(-1) dFMAChR were 40.3%,55.4%and 75.0%,respectively.HE and immunohistochemistry results showed that dFMAChR killed tumor cells by down-regulating the expression of VEGF protein in tumor tissues.CONCLUSION dFMAChR can inhibit the growth of orthotopic xenografts on the human breast cancer by down-regulating the VEGF protein in a dose-dependent manner.
