摘要
目的建立ICR小鼠的焦虑模型,并初步探讨米胚粉对ICR小鼠焦虑症的作用。方法应用不同剂量MCPP建立ICR小鼠焦虑模型,探讨不同剂量米胚粉对ICR焦虑症小鼠的抗焦虑作用,并且排除米胚粉致焦虑作用。结果①采用MCPP建立小鼠焦虑模型。在明暗箱模型中,与生理盐水组相比,不同剂量MCPP均能减少小鼠在明箱停留时间和穿梭次数的趋势,MCPP 1.0 mg·kg^(-1)剂量组在明箱停留时间和穿梭次数与生理盐水组相比都有统计学差异(P<0.05),MCPP 0.25 mg·kg^(-1),0.5 mg·kg^(-1)剂量组仅在穿梭次数与生理盐水组相比有统计学差异(P<0.05);②不同剂量米胚粉对小鼠焦虑行为的影响。与MCPP组1.0 mg·kg^(-1)组相比,米胚粉中剂量组、地西泮组均可以增加小鼠在明箱停留时间,且有统计学差异(P<0.05),但是只有地西泮组的穿梭次数与MCPP组1.0 mg·kg^(-1)组相比有统计学差异(P<0.05);③中剂量+生理盐水组和生理盐水组对明箱停留时间和穿梭次数均无明显差异(P>0.05)。结论经初步ICR小鼠行为学实验检测,中剂量米胚粉能增加小鼠停留在明箱的时间,所以中剂量米胚粉可能有潜在的抗MCPP所致的小鼠焦虑症的作用。
OBJECTIVE To investigate the anti-anxiety effect of rice germ on MCPP treated ICR mice.METHODS ICR mice were administrated with different dose of MCPP and tested in light-dark box to establish anxiety model.Then MCPP treated mice were fed with different dose of rice germ to exam their anti-anxiety effect,and exclude their effect of causing anxiety.RESULTS①The anxiety model of mice was established with MCPP.In the light-dark box experiment,compared to the saline group,different doses of MCPP could reduce the residence time and shuttling times in the open box of mice.Compared to the normal saline group,the residence time and shuttling times in the open box of MCPP 1.0 mg·kg^(-1)group were statistically different(P<0.05),while MCPP 0.25 mg·kg^(-1)group and 0.5 mg·kg^(-1)group only had statistical difference in the number of shuttling(P<0.05).②Effects of different doses of rice germ on anxiety behavior of mice.Compared to the MCPP 1.0 mg·kg^(-1)group,both the medium-dose rice germ group and the diazepam group could statistically increase the residence time in the open box of mice(P<0.05).However,only the shuttling times in the diazepam group had statistical differences compared to the MCPP 1.0 mg·kg^(-1)group(P<0.05).③Effect of medium dose rice germ on normal mice.There was no significant difference in open-box residence time and shuttling times between medium dose+NaCl group and NaCl group(P>0.05).CONCLUSION Based on the preliminary behavioral test of ICR mice,the anxiety model of ICR mice with light-dark box was established.The medium dose of rice rice germ could increase the residence time in the light box.Therefore,the medium dose of rice germ may have the potential effect of anti-MCPP-induced anxiety in mice.
