大果沙棘中黄酮的体外抗炎及抗氧化活性研究 被引量:28

Study on in vitro anti-inflammatory and antioxidant activities of flavonoids from sea buckthorn

摘要 通过采用Griess和酶联免疫吸附测定法检测沙棘黄酮对脂多糖(LPS)诱导小鼠腹腔巨噬细胞(Raw264.7)的细胞吞噬能力、一氧化氮(NO)及炎性因子的影响。分析发现,沙棘黄酮在50~250µg/mL能够显著增加细胞吞噬能力且显著抑制Raw264.7细胞内NO、白细胞介素6(IL-6)、细胞肿瘤坏死因子-α(TNF-α)和环氧化酶-2(COX-2)的产生,并均呈剂量依赖效应。结果表明,沙棘黄酮可通过调控细胞因子和炎性因子分泌而抑制炎症反应。此外,比较了沙棘黄酮与抗坏血酸(VC)的羟自由基、DPPH自由基、ABTS自由基清除能力和总抗氧化(FRAP)差异。结果表明,沙棘黄酮清除DPPH自由基、ABTS自由基的能力和VC相当(IC50值分别为9.014μg/mL、136.86μg/mL),当浓度为80μg/mL时沙棘黄酮的羟自由基清除能力、总还原能力和FRAP分别为59.35%、1.02和2.36mmoL FeSO4/L,说明其具有良好的抗氧化能力。 Griess and enzyme-linked immunosorbent assay were used to detect the effects of sea buckthorn flavonoids on lipopolysaccharide(LPS)-induced mouse peritoneal macrophages(Raw264.7)phagocytic capacity,nitric oxide(NO)and inflammatory factors.Analysis illustrated that sea buckthorn flavonoids at 50-250µg/mL can significantly increase the phagocytic capacity of cells and significantly inhibit the intracellular NO,interleukin 6(IL-6),tumor necrosis factor-α(TNF-α)and Raw264.7 cells.The production of cyclooxygenase-2(COX-2)shows a dose-dependent effect.It showed that sea buckthorn flavonoids can inhibit inflammation by regulating the secretion of cytokines and inflammatory factors.In addition,the differences in hydroxyl free radicals,DPPH free radicals,ABTS free radical scavenging ability and total antioxidant(FRAP)of sea buckthorn flavonoids and ascorbic acid(Vc)were compared.The results showed that the ability of sea buckthorn flavonoids to scavenge DPPH free radicals and ABTS free radicals was equivalent to Vc(IC50 values were 9.014μg/mL,136.86μg/mL,respectively).When the concentration was 80μg/mL,the hydroxyl radical scavenging ability and total reduction capacity and FRAP were 59.35%,1.02and 2.36mmoL FeSO4/L,respectively,indicating that it has good antioxidant capacity.
