摘要
目的探讨过敏性哮喘患儿外周血微小RNA(miR)-145表达及Th1/Th2细胞比例,并分析其相关性。方法选取本院2018年6月至2020年6月门诊及住院收治的过敏性哮喘患儿129例为研究对象,其中轻度组67例,中重度组62例,并选取同期健康体检患儿80名为对照组。辅助性T细胞(Th)1、Th2细胞比例应用流式细胞术检测,血清IgE水平采用全自动生化分析仪检测;第1秒用力呼气容积(FEV1%)水平利用肺功能测定仪检测;对过敏性哮喘患儿进行哮喘控制问卷调查(ACQ),利用实时定量PCR(qRT-PCR)法检测外用白单个核细胞(PBMCs)中miR-145表达情况。结果与对照组比较,轻度组、中重度组患儿FEV1%、PBMCs中Th1细胞亚群比例、Th1/Th2比例及血清IFN-γ水平显著降低(P<0.05),血清IgE、IL-5、IL-13水平及PBCs中miR-145水平、Th2细胞亚群比例显著升高(P<0.05);与轻度过敏性哮喘患儿比较,中重度组过敏性哮喘患儿FEV1%、Th1细胞亚群比例、Th1/Th2比例及血清IFN-γ水平显著降低(P<0.05),血清IgE、IL-5、IL-13水平及PBMCs中miR-145水平、Th2细胞亚群比例显著升高(P<0.05);过敏性哮喘患儿PBMCs中miR-145表达水平与Th1/Th2比例显著负相关(P<0.05)。结论与健康儿童比较,过敏性哮喘患儿外周血PBMCs中miR-145高表达,Th1/Th2比例降低,miR-145可能通过调节Th1/Th2比例影响过敏性哮喘发生。
Objective To investigate the correlation between expression of microRNA(miR)-145 and the proportion of Th1/Th2 cells in peripheral blood of children with allergic asthma.Methods A total of 129 children with allergic asthma admitted to our hospital from June 2018 to June 2020 were selected as the study subjects,including 67 cases in mild group and 62 cases in moderate and severe group,80 children with healthy physical examination in the same period were selected as the control group.Flow cytometry was used to detect the proportions of Th1 and Th2 cells,automatic biochemical analyzer was used to detect level of serum IgE,forced expiratory volume during the first second(FEV1%)was measured by pulmonary function analyzer,asthma control questionnaire(ACQ)was conducted,and real-time quantitative PCR(qRT-PCR)was used to detect the expression of miR-145 in peripheral blood mononuclear cells(PBMCs).Results Compared with the control group,the FEV1%,the percentage of Th1 cell subsets,the ratio of Th1/Th2 in PBMCs and level of interferon(IFN)-gamma in serum of children with allergic diseases in mild group and moderate and severe group were significantly lower(P<0.05),while the levels of IgE,IL-5 and IL-13 in serum,the level of miR-145 and the proportion of Th2 cell subsets in PBMCs were significantly higher(P<0.05).Compared with mild allergic children,the FEV1%,the percentage of Th1 cell subsets,the ratio of Th1/Th2 and level of IFN-gamma in serum of children with allergic diseases in moderate and severe group were significantly lower(P<0.05),while the levels of IgE,IL-5 and IL-13 in serum,the level of miR-145 and the proportion of Th2 cell subsets in PBMCs were significantly higher(P<0.05).The expression of miR-145 in PBMCs of children with allergic asthma was negatively correlated with Th1/Th2 ratio(P<0.05).Conclusion Compared with healthy children,the expression of miR-145 in PBMCs of children with allergic asthma is high and the ratio of Th1/Th2 is decreased,miR-145 may affect the occurrence of allergic asthma by regulating the ratio of Th1/Th2.
