摘要
目的:基于网络药理学探讨柴胡陷胸汤治疗冠状动脉粥样硬化性心脏病(简称冠心病)的作用机制。方法:在中药药理数据库和分析平台(TCMSP)平台筛选出柴胡陷胸汤的主要成分,并在Uni Prot数据库中获取成分相关靶点信息。使用Cytoscape3.8.0构建成分-靶点信息网络图。通过OMIM、Genecards数据库得到冠心病疾病靶点,韦恩图获取柴胡陷胸汤治疗冠心病的共同靶点,String构建蛋白质相互作用网络(PPI)。最后通过R语言程序进行基因本体(GO)生物功能注释,京都基因和基因组数据库(KEGG)通路富集分析。结果:柴胡陷胸汤共筛选出98种成分,与冠心病相关靶点83个,相关性较高的靶点包括肿瘤坏死因子(Tumor Necrosis Factor,TNF)、白细胞介素-6(Interleukin-6,IL-6)、丝裂原活化蛋白激酶1(Mitogen-Activated Protein Kinase 1,MAPK1)、血管内皮生长因子(Vascular Endothelial Growth Factor,VEGF)等,GO主要富集在活性氧类代谢过程、对营养水平的反应、多糖应答、循环系统的血管功能等生物过程,KEGG主要富集在血流剪切力和动脉粥样硬化、缺氧诱导因子-1信号通路、白细胞介素-17(Interleukin-17,IL-17)信号通路、TNF信号通路、Relaxin信号通路等信号通路。结论:柴胡陷胸汤可能通过上述作用机制抑制血管内皮炎症反应,改善心肌缺氧,减缓血栓及斑块形成,从而达到治疗冠心病的功效。
Objective:To explore the mechanism of the Chaihu Xianxiong decoction(柴胡陷胸汤) in treating coronary heart disease based on internet pharmacology.Methods:The main components of the Chaihu Xianxiong decoction were selected on the platform of TCMSP,and the related target information was obtained in Uniprot database.Construct component-target information network diagram using cytoscape 3.8.0.Through OMIM and Genecards Database,we got the target of coronary heart disease,and Venn diagram got the common target of the Chaihu Xianxiong decoction for coronary heart disease,String constructed PPI network.Finally,GO bio-functional annotation and KEGG pathway enrichment analysis were carried out by R language program.Results:A total of 98 components of the Chaihu Xianxiong decoction were screened out.There were 83 targets related to coronary heart disease.The targets with high correlation included TNF,IL-6,MAPK1 and VEGFA,go is mainly enriched in biological processes such as reactive oxygen species metabolism,response to nutritional level,polysaccharide response,vascular function of circulatory system,etc..KEGG is mainly concentrated in the blood flow shear stress and arteriosclerosis,HIF-1 signal pathway,IL-17 signal pathway,TNF signal pathway,Relaxin signal pathway and so on.Conclusion:The Chaihu Xianxiong decoction may inhibit the inflammatory reaction of vascular endothelium,improve myocardial hypoxia,slow down the formation of thrombus and plaque,and thus achieve the effect of treating coronary heart disease.
