摘要
目的探讨高龄肺腺癌患者癌组织内转化生长因子-β激活的长链非编码RNA(lncRNA-ATB)、微小RNA34a(miR-34a)和微小RNA-145(miR-145)的表达水平与其生存情况的关系。方法选取2014年8月至2016年8月在该院治疗的112例高龄肺腺癌患者为研究对象,采用qRT-PCR检测患者癌组织及癌旁组织中lncRNA-ATB、miR-34a和miR-145表达水平,分别根据lncRNA-ATB、miR-34a和miR-145的表达水平将患者分为高、低表达组,以总生存率(OS)作为检测指标,采用Kaplan-Meier法分别分析两组患者的预后情况,并采取Cox法探讨影响患者预后的独立危险因素。结果肺腺癌组织lncRNA-ATB表达水平显著高于癌旁组织,肺腺癌组织miR-34a和miR-145表达水平显著低于癌旁组织,差异均有统计学意义(P<0.05)。随访发现共24例患者死亡,所有患者的3年OS为78.57%。其中lncRNA-ATB高、低表达组3年OS分别为71.43%、85.71%;miR-34a高、低表达组3年OS分别为91.07%、66.07%;miR-145高表达组高、低表达组3年OS分别为89.29%、67.86%,Log-rank检验显示,各指标高、低表达患者的OS比较,差异均有统计学意义(P<0.05)。影响患者预后的多因素Cox分析结果显示,肿瘤TNM分期升高、发生淋巴结转移和胸膜侵犯、lncRNA-ATB高表达以及miR-34a、miR-145低表达是影响患者预后的独立危险因素(P<0.05)。结论lncRNA-ATB在高龄肺腺癌患者癌组织中呈高表达,miR-34a、miR-145在高龄肺腺癌患者癌组织中呈低表达,且lncRNA-ATB高表达,miR-34a、miR-145低表达是影响患者预后的独立危险因素。
Objective To investigate the levels of long non-coding RNA-activated by transforming growth factorβ(lncRNA-ATB),microRNA(miR)-34a and miR-145 in tissues of elderly patients with lung adenocarcinoma and their correlation with survival.Methods A total of 112 elderly patients with lung adenocarcinoma treated in the hospital from August 2014 to August 2016 were selected as the research subjects,the lncRNA-ATB,miR-34a and miR-145 levels in the pathological tissues and tissues adjacent to carcinoma of the patients were detected by qRT-PCR,the patients were divided into high and low expression groups according to the expression levels of lncRNA-ATB,miR-34a and miR-145,and the overall survival rate(OS)was used as the detection index.Kaplan-Meier method was used to analyze the two groups of patients.The prognosis and Cox method were used to explore the independent factors affecting the prognosis of patients.Results The expression levels of lncRNA-ATB in lung adenocarcinoma tissues were significantly higher than those in adjacent tissues.The expression levels of miR-34a and miR-145 in lung adenocarcinoma tissues were significantly lower than those in adjacent tissues,and the differences were statistically significant(P<0.05).A total of 24 patients died during follow-up and the 3-year OS of all patients was 78.57%.Among them,the lncRNA-ATB high and low expression groups had 71.43%and 85.71%of 3-year OS;miR-34a high and low expression groups had 91.07%and 66.07%of 3-year OS;miR-145 high and low expression groups had 89.29%and 67.86%of 3-year OS,respectively.The differences in OS between the high and low expression groups of the three indicators of Log-rank test were statistically significant(P<0.05).Multivariate analysis of Cox showed that elevated tumor TNM stage,lymph node metastasis and pleural invasion occurred,high expression of lncRNA-ATB,and low expression of miR-34a and miR-145 were independent risk factors for the prognosis(P<0.05).Conclusion lncRNA-ATB is highly expressed in lung adenocarcinoma tissues of elderly patients,miR-34a and miR-145 are lowly expressed in lung adenocarcinoma tissues of elderly patients,and highly expressed lncRNA-ATB,lowly expressed miR-34a and miR-145 are independent risk factors that affect the prognosis of patients.
