摘要
目的探讨核苷酸寡聚化结构域(NOD)样受体蛋白3(NLRP3)及其相关炎症因子IL-1β和IL-18在神经病理性疼痛(NP)小鼠的表达。方法 36只小鼠分为模型组(建立慢性压迫性神经损伤模型,18只)和假手术组(18只)。于术前和术后3、7、14、21和28 d,观察小鼠行为学表现,采用实时定量PCR和Western blot法分别检测小鼠背根神经节组织NLRP3 mRNA和蛋白表达,ELISA法检测IL-1β和IL-18的表达。结果与假手术组比较,模型组在术后3、7、14、21和28 d机械缩足反射阈值降低,热缩足反射潜伏期缩短(P<0.05),术后3、7和14 d NLRP3 mRNA和蛋白表达升高(P<0.05),术后3、7、14、21、28 d IL-1β和IL-18表达升高(P<0.05)。结论 NLRP3及其相关炎症因子IL-1β和IL-18在NP小鼠表达升高,可能在NP的发生、发展中起到重要作用。
Objective To investigate the expressions of NOD like receptor protein 3(NLRP3) and its relevant inflammatory factors IL-1β and IL-18 in the mice with neuropathic pain(NP).MethodsA total of 36 mice were divided into two groups with 18 mice each.Mice models with NP were established in group A.The mice in group B were taken as sham operation controls.The behavioral performance of mice was observed before and 3,7,14,21 and 28 days after operation.At the same time points, the mRNA and protein expressions of NLRP3 were detected by real-time quantitative PCR and Western blot, and the expressions of IL-1β and IL-18 were measured by ELISA.Results The paw withdrawal threshold was lower and paw withdrawal latency was shorter in group A than those in group B on the 3^(rd),7^(th),14^(th),21^(st) and 28^(th) day after operation(P<0.05).The mRNA and protein expressions of NLRP3 were higher in group A than those in group B on the 3^(rd),7^(th) and 14^(th) day after operation(P<0.05).The expressions of IL-1β and IL-18 were higher in group A than those in group B on the 3^(rd),7^(th),14^(th),21^(st) and 28^(th) day after operation(P<0.05).Conclusion The expressions of NLRP3 and its related inflammatory factors IL-1β and IL-18 in NP mice are increased, which may play an important role in the occurrence and development of NP.
