摘要
目的探讨芪苈强心胶囊对心力衰竭大鼠心室重构及缺氧诱导因子1(HIF-1)和血管内皮生长因子(VEGF)表达的影响。方法结扎冠状动脉成功建立心肌梗死后心力衰竭大鼠模型32只,分为模型对照(模型组,14只)和中药处理(中药组,18只)两组;另取20只大鼠为假手术组。中药组给予芪苈强心胶囊1 g·kg^(-1)·d^(-1)灌胃治疗6周。6周后检测三组血流动力学指标,计算左心室重量指数(LVMI)。采用心脏超声心动图检测三组大鼠左心室舒张末内径(LVEDD)、左心室收缩末内径(LVESD)、室间隔厚度(IVS)及左心室射血分数(LVEF)。采用RT-PCR及ELISA法检测大鼠HIF-1和VEGF的表达。结果与假手术组比较,中药组和模型组大鼠左心室收缩压、左心室内压上升最大速率及左心室内压下降最大速率均降低,左心室舒张期末压升高(P<0.01),IVS、LVEDD、LVESD和LVMI增加,LVEF降低(P<0.05),HIF-1和VEGF浓度及其mRNA相对表达量增加(P<0.01)。与模型组比较,中药组可以逆转上述指标的变化(P<0.05或P<0.01)。结论芪苈强心胶囊能改善心肌梗死后心力衰竭大鼠心功能,逆转心室重构;其机制可能与提高HIF-1和VEGF的表达及促进血管新生有关。
Objective To investigate the effects of Qili qiangxin on ventricular remodeling and the expressions of hypoxia-inducible factor 1(HIF-1)and vascular endothelial growth factor(VEGF)in SD rats with heart failure.Methods The rat models with heart failure after myocardial infarction were established by ligating the coronary arteries,which then were divided into two groups of A(model control,14 rats)and B(treated with Qili qiangxin 1 g·kg^(-1)·d^(-1) intragastrically for 6 weeks,18 rats).Another 20 SD rats were sham operated as the controls(group C).Six weeks later,the hemodynamic indexes were measured and left ventricular mass index(LVMI)was calculated.The left ventricular end-diastolic diameter(LVEDD),left ventricular end-systolic diameter(LVESD),ventricular septal thickness(IVS)and LVEF were detected by echocardiography.The expressions of HIF-1 and VEGF were detected by RT-PCR and ELISA.Results Compared with group C,the left ventricular systolic pressure,the maximum rate of left ventricular pressure increase,the maximum rate of left ventricular pressure and LVEF were decreased,and the left ventricular end diastolic pressure,IVS,LVEDD,LVESD,LVMI,the concentrations and mRNA relative expressions of HIF-1 and VEGF were increased in groups of A and B(P<0.05 or P<0.01).Group B can reverse the changes of above indicators in group A(P<0.05 or P<0.01).Conclusion Qili qiangxin can improve the heart function of the rats with heart failure after myocardial infarction and reverse ventricular remodeling,which may be related to the increases of HIF-1 and VEGF expressions and the promotion of angiogenesis.
