摘要
[目的]探讨牡丹皮乙醇提取物(MCE)对1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)诱导的帕金森病模型小鼠多巴胺能神经元损伤的调控作用.[方法]将C57/BL6小鼠随机分为对照组、模型组和MCE干预组,其中MCE低、中、高剂量组分别灌胃给予25、50、100 mg/kg MCE.运用神经行为学、免疫组织化学和Western blot技术检测MCE对MPTP诱导的帕金森病模型小鼠行为学变化、黑质致密部多巴胺能神经元缺失及线粒体复合物活性蛋白的影响.[结果]与模型组比较,MCE各剂量组小鼠运动能力明显得到改善(P<0.05),MCE中、高剂量组酪氨酸羟化酶阳性细胞数明显增多(P<0.01),各种蛋白表达水平明显升高.[结论]MCE可抑制MPTP诱导的帕金森病模型小鼠线粒体功能障碍,具有神经保护作用.
OBJECTIVE To investigate the regulating effects of ethanol extract of Cortex Moutan(MCE)on dopaminergic neuronal damage in Parkinson’s disease mouse model induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP).METHODS C57/BL6 mice were randomly divided into control group,model group,and MCE intervention groups.Low,medium and high dose MCE groups were given 25,50 and 100 mg/kg MCE by gavage respectively.Neurobehavioral,immunohistochemical and western blot methods were used to detect the effects of MCE on behavioral changes,dopaminergic neuron loss in substantia nigra pars compacta and active protein of mitochondrial complex in Parkinson’s disease model mice induced by MPTP.RESULTS Compared with the model group,the motor ability of mice in MCE groups was significantly improved(P<0.05).The number of tyrosine hydroxylase positive cells increased significantly in MCE medium and high dose group(P<0.01),and the expression levels of various proteins were significantly increased.CONCLUSION MCE can inhibit mitochondrial dysfunction in Parkinson’s disease model mice induced by MPTP and has neuroprotective effect.
