摘要
前期研究发现,大豆GmSARK基因编码一个典型的LRR型类受体蛋白激酶,并且为叶片衰老的正调控因子.构建了LRR结构域缺失型GmSARK(即GmSARK^(⊿LRR)),并通过农杆菌介导的遗传转化方法获得了3个诱导型启动子GVG驱动GmSARK^(⊿LRR)过表达的转基因拟南芥独立株系.衰老相关的表型分析和分子鉴定结果表明,GVG:GmSARK^(⊿LRR)丧失早衰表型,暗示着LRR基序在与配基结合和衰老信号传递中起重要作用.本论文的研究结果为深入解析SARK介导的衰老信号通路奠定了重要基础.
Our group previously found that GmSARK encodes a typical leucine-rich repeat receptor-like protein kinase(LRR-RLK)and functions as a positive regulator of leaf senescence.In the current study,we constructed a truncated GmSARK lacking the LRR,designated as GmSARK^(⊿LRR),and obtained three independent lines of GmSARK^(⊿LRR) overexpressing Arabidopsis driven by an induced promoter GVG.The subsequent phenotypic analysis and molecular assays demonstrated that loss of LRR region totally inhibited GmSARKinduced precocious leaf senescence.Our results implied that the LRR motifs may be import for the ligand binding and the signal transduction pathway of GmSARK-induced precocious leaf senescence.The current study laid foundation for a deeper understanding towards SARK-induced leaf senescence signaling.
