Assisted Breeding Technology of SSR Markers on Hybrid Soybean

摘要 基于鉴定真假大豆杂交种需要,课题组系统研究掌握了SSR分子标记鉴定技术,该研究通过SSR分子标记鉴定技术来辅助了杂交种优势豆-A-5的选育。其中优势豆-A-5扩增的互补谱带的9对SSR分子标记分布于7个连锁群,用9对标记检测杂交种纯度,最后检测合格的种子进行品种审定的比较试验、区域试验、生产试验。该选题对大豆杂交种育种有较好的辅助作用,提供了试验参考。优势豆-A-5是利用大豆质核互作雄性不育"三系法"选育的大豆杂交种,不育系为JLSXCMS1,恢复系为中119-99,2019年通过审定(晋审豆20190002),2014-2015年在山西春播中晚熟区区域试验平均产量3570 kg/hm^(2),比对照‘晋豆19号’增产13.5%;2016年生产试验,优势豆-A-5平均产量3217.5 kg/hm^(2),比对照‘晋豆19号’增产12.1%。粗蛋白含量为42.10%、粗脂肪含量为20.96%,适宜在山西大豆春播中晚熟区和南部夏播区种植。2020年在山西运城市推广示范,产量3675 kg/hm^(2)。 Based on the need of identifying true and false soybean hybrids,the biotechnology of SSR marker are used to guide the breeding of hybrid soybean.The research group has systematically studied and mastered the SSR molecular marker identification technology of soybean hybrids.The technique assisted the breeding of YoushidouA-5,9 pairs of SSR markers about Youshidou-A-5 amplified complementary band were distributed in seven linkage groups.The purity of hybrid soybean was detected by 9 pairs of SSR markers.The qualified seeds are tested in the comparative test,the regional test and the production test.This topic has a good assisting effect on the breeding of hybrid soybean,and provides experimental reference.Youshidou-A-5 is soybean hybrid which was selected by the ’three-line’ method of the cytoplasmic-nuclear male sterile line.The maternal sterile line is JLSXCMS1,and the parental restorer line is Zhong119-99,it was approved in 2019(Authorized number:Jinshendou-20190002).During the ye ar of 2014 to 2015 regional test of medium-late maturity-spring sowing soybean of Shanxi,the average yield was 3 570 kg/hm^(2),increased 13.5% more than that of ’Jindou19’(CK).The average yield was 3 217.5 kg/hm^(2),increased 12.1% more than that of ’Jindou19’(CK) in production test in 2016.Youshidou-A-5 seed protein content is 42.10%,fat content is 20.96%.The variety is adapted to be cultivated in medium-late maturing areas of spring sowing soybean in Shanxi,and the south summer sowing areas of Shanxi.In 2020,it was promoted and demonstrated in Yuncheng City of Shanxi Province,the yield was 3 675 kg/hm^(2).
