甘蓝型油菜显性核不育全不育系高纯度制种技术分析 被引量:1

Analysis on High-purity Seed Production Technology of Dominant Genic Male Sterile(DGMS) Full Sterile Line in Rapeseed

摘要 本研究开展了甘蓝型油菜显性核不育全不育系高纯度制种技术的研究,利用离体培养、无性扩繁和BE10分子标记辅助选择等技术繁殖纯合型不育株。结果表明,利用幼嫩枝条诱导获得试管苗不容易出现混杂,基因型鉴定更为简单、准确。通过对试管苗移栽技术的研究发现,培养土移栽试管苗可有效提高成活率,两次移栽的最佳时期为9月底至10月初。与原有产量585 kg/hm^(2)相比,利用显性核不育无性扩繁技术生产全不育系产量为827 kg/hm^(2),比原有产量提高了40%,田间育性鉴定结果显示全不育系纯度可达99%以上,为进一步推动甘蓝型油菜显性核不育系的应用奠定技术基础。 We studied the high-purity seed production technology of DGMS full sterile line in Brassica napus L..The homozygous sterile lines were propagated by in vitro culture,asexual propagation,and BE10 molecular marker-assisted selection.The results showed that it was not easy to mix genotypes when young shoots are used to induce sterile lines in vitro,and genotype identification was more simple and accurate.Results of the study on the transplanting technology of plantlets in vitro suggested that culture soil for transplantation could effectively improve the survival rate of plantlets.The best time for two transplantings was from the end of September to the beginning of October.The yields of DGMS full sterile lines obtained by using asexual propagation technology were827 kg/hm^(2),increased by 40% compared with previous yields of 585 kg/hm^(2).Field identification result showed the purity of full sterile line was over 99%.The results provide a technical foundation to further promote the application of DGMS in Brassica napus L..
