基于网络药理学探讨穿心莲治疗新型冠状病毒肺炎机制研究 被引量:4

Study on the Mechanism of Using Andrographis Herba in the Treatment of Novel Coronavirus Pneumonia Based on Network Pharmacology

摘要 目的:通过网络药理学方法探索穿心连治疗新型冠状病毒肺炎的作用靶点及分子机制。方法:通过中药系统药理学分析平台(TCMSP)数据库筛选穿心莲的活性成分及相关的药物靶点,同时在GeneCards数据库筛选出新型冠状病毒肺炎的相关疾病靶点,交集基因即为穿心莲治疗新冠肺炎的核心靶点。随后利用Cytoscape3.7.2软件和String数据库构建药物活性成分-靶点网络和蛋白质相互作用关系的网络图。最后通过DAVID数据库对交集靶点进行GO功能富集和KEGG通路富集分析,并构建成分-靶点-通路图。结果:筛选出穿心莲的活性成分24个及作用靶点53个,新型冠状病毒肺炎的靶点259个,交集靶点14个。KEGG富集分析涉及通路17条,主要包括癌症、细胞凋亡、肌萎缩性侧索硬化症(ALS)、MAPK信号通路、神经营养蛋白信号通路等。结论:穿心莲中的多种活性成分可以通过介导多靶点、多通路发挥治疗新型冠状病毒肺炎的作用。 Objective:To explore the target and molecular mechanism of using Andrographis Herba in the treatment of novel coronavirus pneumonia through network pharmacology.Methods:The active ingredients and related ingredient targets of Andrographis Herba were screened through the TCMSP database,and the novel coronavirus pneumonia related targets were searched in the GeneCards database.The intersection of the two genes is the core target of Andrographis Herba in the treatment of novel coronary pneumonia.Then use Cytoscape 3.7.2 software and String database to construct a network diagram of the active ingredient-target network and protein-protein interaction relationship network.Finally,the gene ontology(GO)enrichment and KEGG pathway enrichment analysis were performed on the intersection targets through the DAVID database and finished the components-targets-pathway network.Results:24 active components of Andrographis Herba and 53 targets were selected,and 259 targets of novel coronavirus pneumonia.There were only 14 intersection targets.KEGG enrichment analysis involves 17 pathways,involving in cancer pathway,apoptosis,amyotrophic lateral sclerosis(ALS),MAPK signaling pathway,and neurotrophin signaling pathway.Conclusion:Andrographis Herba exerts clinical effects on novel coronavirus pneumonia through multiple active ingredients,multiple targets,and multiple pathways.
