摘要
目的:探讨紫草素调控TGF-β1/Survivin通路对银屑病皮肤炎症反应的影响。方法:选择2019年8月~2020年8月由我院诊治的银屑病患者95例为实验组皮肤组织标本,30例正常健康人群外科手术切除多余皮肤组织标本,给予实验组紫草素治疗。记录患者治疗前后的PADI积分、组织中TGF-β1/Survivin的表达及双荧光酶实验结果。结果:TGF-β1阳性定位于银屑病皮损表皮全层或中下层的角质形成细胞胞核及胞浆内,对照组中仅少数基底层细胞呈阳性表达;Survivin阳性定位于细胞浆、胞核,在银屑病皮损的表皮角质形成细胞各层和中下层角质形成细胞中呈不同程度的阳性表达,在对照组皮损中几乎不表达。治疗后实验组PASI评分低于治疗前(P<0.05);对照组TGFβ1、Survivin灰度值均低于实验组(治疗前、后),治疗后实验组TGFβ1、Survivin灰度值低于治疗前实验组(P<0.05);TGFβ1/Survivin在银屑病皮损组织中表达下调;刺激Hacat细胞后,TGFβ1/Survivin的表达下调;TGFβ1/Survivin在银屑病皮损组织中表达较a、b图有明显上升(P<0.05)。结论:紫草素可通过调控TGF-β1/Survivin通路改善银屑病患者的皮肤炎症反应。
Objective:To investigate the effect of Shikonin on skin inflammatory response for patients with psoriasis through regulating TGF-β1/Survivin pathway.Methods:The skin tissues removed from 95 patients with psoriasis and 30 healthy individuals form August 2019 to August 2020 were collected,and set as an experimental group and a controlled group,respectively.The experimental group received Shikonin treatment.PADI score,expression of TGF-β1/Survivin in tissues and the double fluorescent enzyme test results were recorded before and after treatment.Results:In the experimental group,positive TGF-β1 was found in the nucleus and cytoplasm of keratinocytes in the whole or middle lower epidermis of psoriasis.In the controlled group,only a few basal cells showed positive expression of TGF-β1;Survivin was localized in the cytoplasm and nucleus,and was expressed in different degrees in the keratinocytes of the epidermis of psoriasis.Survivin was almost not expressed in the controlled group.PASI score was decreased in the experimental group after treatment(P<0.05).The gray values of TGF-β1 and Survivin in the controlled group were lower than those in the experimental group,while the gray values of TGF-β1 and Survivin after treatment of the experimental group were lower than before treatment(P<0.05).The expression of TGF-β1/Survivin was down regulated in psoriatic lesions.After stimulation of HaCaT cells,the expression of TGF-β1/Survivin was down regulated,TGF-β1/Survivin expression in psoriasis lesions was significantly higher than that in images A and B(P<0.05).Conclusion:Shikonin can improve the skin inflammation in patients with psoriasis by regulating the TGF-β1/Survivin pathway.
