辽东栎白粉病叶片的菌群结构和多样性分析

摘要 为明确辽东栎白粉病叶片的菌群结构和多样性,利用Illumina MiSeq技术检测细菌16S rDNA和真菌ITS序列,分析辽东栎白粉病叶片的细菌和真菌类群。在门的水平上,辽东栎白粉病叶片细菌的优势菌群为变形菌门(Proteobacteria)、拟杆菌门(Bacteroidetes)、放线菌门(Actinobacteria);真菌的优势菌群为子囊菌门(Ascomycota)和担子菌门(Basidiomycota)。在属的水平,细菌的优势菌属为鞘脂单胞菌属(Sphingomonas)、假单胞菌属(Pseudomonas)、薄层菌属(Hymenobacter)、甲基杆菌属(Methylobacterium)、金色单胞菌属(Aureimonas)、马赛菌属(Massilia);真菌的优势菌属为白粉菌属(Erysiphe)、球腔菌属(Mycosphaerella)、短梗霉属(Aureobasidium)。Alpha多样性分析显示,真菌菌群的Chao1指数大于细菌,细菌菌群的Shannon指数和Simpson指数大于真菌,表明检测到的真菌总数比细菌多,而细菌菌群的多样性、丰富度和均匀度高于真菌。研究结果为进一步探究柞树白粉病致病菌群及发病机制提供了理论依据。 To understand the flora structure and diversity of Quercus wutaishanica leaves infected with powdery mildew, bacterial 16 S rDNA and fungal ITS were sequenced based on Illumina MiSeq technology and were used to analyze the bacterial and fungal flora of Q. wutaishanica leaves infected with powdery mildew. At the phylum level, the dominant bacterial flora were Proteobacteria, Bacteroidetes and Actinobacteria, whereas the dominant fungal flora were Ascomycota and Basidiomycota. At the genus level, the dominant bacteria were Sphingomonas, Pseudomonas, Hymenobacter, Methylobacterium, Aureimonas and Massilia, while the dominant fungi were Erysiphe, Mycosphaerella and Aureobasidium. The results of alpha diversity analysis showed that the Chao1 index of fungal flora was higher than that of bacteria, and the Shannon index and Simpson index of bacterial flora were higher than those of fungi, indicating that the total number of fungi detected was more than that of bacteria, while the diversity, abundance and evenness of bacterial flora were higher than those of fungi. The results of this study provide a theoretical basis for further exploring the pathogenic flora and pathogenesis of powdery mildew of oaks.
