摘要
长链非编码RNA(long non-coding RNA,LncRNA)是一类转录本长度大于200 nt但无蛋白质编码能力的基因组转录产物。研究发现,LncRNA可以在转录前、转录及转录后水平调控基因表达,参与表观遗传修饰、X染色体剂量补偿及基因组印记等事件。胚胎发育是一个复杂且有序的过程,其受一系列事件影响,如母系到合子的过渡(maternal-to-zygotic transition,MZT)和合子基因组激活过程(zygotic genome activation,ZGA)等。目前研究认为LncRNA可以通过参与卵母细胞成熟与精子发生、ZGA以及调控胚胎干细胞(Embryonic stem cells,ESCs)多能性等过程影响胚胎发育。综述了LncRNA在胚胎发育中的机制研究进展,为全面解析植入前胚胎中LncRNA异质性的产生机制及其与细胞命运决定的关联奠定基础。
Long non-coding RNA(LncRNA)is a class of genomic transcripts with a transcript length of more than 200 nt but no protein-coding ability.Studies have shown that LncRNA can regulate gene expression at pre-transcriptional,transcriptional and posttranscriptional levels,and participate in epigenetic modification,X chromosome dose compensation and genomic imprinting.Embryonic development is a complex and orderly process,which is affected by a series of events,such as maternal to zygotic transition(MZT)and zygotic genome activation(ZGA).Current studies suggest that LncRNA can affect embryonic development by participating in oocyte maturation,spermatogenesis,ZGA,and regulating the pluripotency of Embryonic stem cells(ESCs).The research progress on the mechanism of LncRNA in embryonic development was reviewed,which laid the foundation for a comprehensive analysis of the mechanism of LncRNA heterogeneity in preimplantation embryos and its association with cell fate determination.
