摘要
目的:探讨姜黄素(curcumin)对A549肺癌细胞增殖、凋亡和迁移能力以及YES相关蛋白1(YAP1)表达的影响。方法:采用CCK8法检测姜黄素对A549细胞的IC50浓度;分别用10μmol/L、20μmol/L、40μmol/L姜黄素处理A549肺癌细胞(Cur10组、Cur20组和Cur40组),CCK-8法、流式细胞术及Transwell小室实验检测姜黄素对A549细胞增殖、凋亡及迁移能力的影响;采用WesternBlot检测姜黄素对YAP1蛋白表达影响。结果:姜黄素处理后,Cur10组、Cur20组和Cur40组细胞的增殖能力均显著低于BC组,增殖抑制率和凋亡率逐渐升高,迁移能力逐渐降低,且呈剂量依赖性;与对照组相比,Cur10组、Cur20组和Cur40组YAP1蛋白表达水平均显著降低,且Cur20组和Cur40组显著低于Cur10组,Cur40显著低于Cur20组。结论:姜黄素可以呈剂量依赖性抑制A549肺癌细胞增殖和迁移能力,促进其凋亡,其机制可能与对YAP1蛋白的抑制有关。
Objective:To investigate the effect of curcumin on the proliferation,apoptosis,migration and expression of YES related protein 1(YAP1)in A549 lung cancer cells.Methods:CCK8 was used to detect IC50 concentration of curcumin on A549 cells.A549 lung cancer cells(Cur10 group,Cur20 group and Cur40 group)was treated with 10,20,40μmol/L curcumin,respectively.CCK-8 method,flow cytometry and Transwell chamber experiment were used to detect the effects of curcumin on the proliferation,apoptosis and migration of A549 cells.Western Blot was used to detect the effect of curcumin on YAP1 protein expression.Results:After curcumin treatment,the proliferation ability of Cur10 group,Cur20 group and Cur40 group was significantly lower than that of BC group.The proliferation inhibition rate and apoptosis rate gradually increased,and the migration ability gradually decreased in a dose-dependent manner.Compared with the control group,the YAP1 protein expression levels in the Cur10 group,Cur20 group and Cur40 group were significantly reduced,and the Cur20 group and Cur40 group were significantly lower than the Cur10 group,and the Cur40 group was significantly lower than the Cur20 group.Conclusion:Curcumin can inhibit the proliferation and migration of A549 lung cancer cells in a dose-dependent manner,and promote their apoptosis.The mechanism may be related to the inhibition of YAP1 protein.
