摘要
目的探讨咖啡因对糖尿病小鼠肠道菌群的影响。方法选取25只SPF级BALB/c雄性小鼠,利用链脲霉素诱导糖尿病小鼠模型,随机选取20只,分为咖啡因组和非咖啡因组,每组10只。咖啡因组给予10 mg/(kg·d)的咖啡因灌胃,非咖啡因组给予等量生理盐水灌胃。观察4周后2组小鼠空腹血糖,取2组小鼠的新鲜粪便样本,采用16S rDNA高通量测序技术分析肠道菌群的变化。结果咖啡因组与非咖啡因组小鼠血糖、肠道菌群α多样性、β多样性和菌群在纲、目水平上的相对丰度比较差异无统计学意义(P>0.05)。咖啡因组菌群在科、属水平上乳杆菌的相对丰度明显高于非咖啡因组(P<0.05)。结论咖啡因的连续摄入对糖尿病小鼠的血糖无明显不利影响,摄入咖啡因能显著提升糖尿病小鼠肠道中乳杆菌的丰度。
Objective To explore the effect of caffeine on intestinal flora in mice with diabetes.Methods Twenty five SPF-grade BALB/c male mice were selected,and then streptozotocin(STZ)was used to induce diabetic mouse models,and then 20 diabetic mice were randomly selected and divided into caffeine group(n=10)and non-caffeine group(n=10).Caffeine group was given 10 mg/(kg·d)caffeine by gavage,while non-caffeine group was given the same volume of normal saline by gavage.Levels of fasting blood glucose were measured after treatment for 4 weeks in two groups.Fresh fecal samples were taken,and 16 S rDNA high-throughput sequencing technology was used to analyze the changes of intestinal flora in mice with diabetes in two groups.Results No significant differences were found in blood glucose,αdiversity andβdiversity of intestinal flora,and relative abundance of flora at the levels of class and order between caffeine group and non-caffeine group(P>0.05).The relative abundance values of lactobacilli at the levels of family and genus in caffeine group were significantly higher than those in non-caffeine group(P<0.05).Conclusion The continuous intake of caffeine has no obvious adverse effect on the blood sugar of diabetic mice,and the intake of caffeine may significantly increase the abundance of lactobacilli in the intestine of diabetic mice.
