肝癌组织中SOCS1、GSTP1及DKK3基因甲基化状态及与临床病理的关系 被引量:3

Methylation Conditions of SOCS1,GSTP1 and DKK3 Genes in Liver Cancer Tissues and Their Relationships with Clinical Pathology

摘要 目的分析肝癌组织中细胞因子信号转导抑制因子1(SOCS1)、谷胱甘肽S-转移酶P1(GSTP1)及Dickkopf相关蛋白3(DKK3)基因甲基化状态及与临床病理的关系。方法取2015年1月—2019年12月在本院接受手术治疗的肝癌患者的癌组织及癌旁组织各70例。另选取同期在本院接受肝胆外科手术术后病理证实为正常肝组织20例。比较肝癌组织、癌旁组织、正常肝组织SOCS1、GSTP1、DKK3基因甲基化状态及不同临床病理的肝癌组织SOCS1、GSTP1、DKK3基因甲基化状态,分析肝癌组织SOCS1、GSTP1、DKK3基因甲基化状态与临床病理的关系。结果正常肝组织未见SOCS1、GSTP1、DKK3基因甲基化现象;肝癌组织SOCS1、GSTP1、DKK3基因甲基化阳性率显著高于癌旁组织(P<0.01);乙型肝炎表面抗原(HBsAg)阳性肝癌患者SOCS1基因甲基化阳性率显著高于HBsAg阴性患者,有血管侵犯的肝癌组织GSTP1基因甲基化阳性率显著高于无血管侵犯的肝癌组织,有肝硬化的肝癌组织DKK3基因甲基化阳性率显著高于无肝硬化的肝癌组织(P<0.05,P<0.01);肝癌组织SOCS1基因甲基化与HBsAg呈正相关、GSTP1基因甲基化与血管侵犯呈正相关、DKK3基因甲基化与肝硬化呈正相关(P<0.05,P<0.01)。结论肝癌组织及癌旁组织均存在SOCS1、GSTP1、DKK3基因甲基化现象,且肝癌组织SOCS1、GSTP1、DKK3基因甲基化阳性率高于癌旁组织,并分别与HBsAg、血管侵犯、肝硬化密切相关。 Objective To analyze methylation conditions of suppressor of the cytokine signaling 1(SOCS-1),glutathione S-transferase P1(GSTP1)and Dickkopf-related protein-3(DKK3)in liver cancer tissues and their relationships with clinical pathology.Methods Cancer tissues and adjacent tissues of 70 patients with liver cancer who had undergone surgical treatments between January 2015 and December 2019 were collected,and other 20 normal liver tissues confirmed by postoperative pathological results were collected from patients undergoing surgeries in Department of Hepatobiliary Surgery at the same period were selected.Methylation conditions of SOCS1,GSTP1 and DKK3 genes in liver cancer tissues,adjacent tissues and normal liver tissues,as well as in liver cancer tissues with different clinical pathological results were compared.The relationships between methylation conditions of SOCS1,GSTP1 and DKK3 genes in liver cancer tissues with clinical pathology were analyzed.Results No methylation of SOCS1,GSTP1 or DKK3 genes was observed in normal liver tissues.The positive rates of methylation conditions of SOCS1,GSTP1 and DKK3 genes in liver cancer tissues were significantly higher than those in adjacent tissues(P<0.01).The positive rate of methylation of SOCS1 gene in hepatitis B surface antigen(HbsAg)-positive tissues was significantly higher than that in HbsAg-negative tissues.The positive rate of methylation of GSTP1 gene in liver cancer tissues with vascular invasion was significantly higher than that in liver cancer tissues without vascular invasion,and positive rate of methylation of DKK3 gene in liver cancer tissues with liver cirrhosis was significantly higher than that in liver cancer without liver cirrhosis(P<0.05,P<0.01).Methylation of SOCS1 gene in liver cancer tissues positively correlated with HbsAg,and methylation of GSTP1 gene positively correlated with vascular invasion,and methylation of DKK3 gene positively correlated with liver cirrhosis(P<0.05,P<0.01).Conclusion Methylation conditions of SOCS1,GSTP1 and DKK3 genes in liver cancer tissues and adjacent tissues can been both found,and positive rates of methylation of SOCS1,GSTP1 and DKK3 genes in liver cancer tissues are higher than those in adjacent tissues,and it is closely related to HbsAg,vascular invasion and liver cirrhosis.
