Feasibility of Immune Regulation by Xiaoyao Pills to Psychological Stress-induced Infertility and Its Potential Mechanism Based on Network Pharmacology

摘要 目的应用网络药理学探讨逍遥丸对心理应激性不孕免疫调节的可行性和潜在作用机制。方法利用中药系统药理学数据库和分析平台(TCMSP)筛选逍遥丸具有较高活性的化合物成分以及对应的靶标蛋白,通过Universal Protein数据库查询靶标蛋白的基因名称,选择生物物种为Homo sapiens。在Online Mendelian Inheritance in Man和Genecards两个数据库查找相关的基因靶点。使用蛋白质互相作用平台(STRING)构建蛋白相互作用网络(PPI)以筛选重要靶点。使用R语言cluster Profiler软件包,对共同靶点进行GO和KEGG功能富集分析。结果逍遥丸、心理应激性不孕和免疫调节三者交集有53个共同靶点,PPI网络拓扑分析相关靶点有白细胞介素(IL)6、信号转导与转录激活因子3、肿瘤坏死因子(TNF)、IL1β、丝裂原活化蛋白激酶1等共50个。通过富集分析信号通路主要集中在IL17、TNF信号通路等。结论逍遥丸可能通过IL17、TNF信号通路对心理应激性不孕进行免疫调节,为后续的实验研究和临床应用提供理论依据。 Objective To investigate feasibility and of immune regulation by Xiaoyao Pill on psychological stress-induced infertility and its potential mechanism based on network pharmacology.Methods Traditional Chinese medicine systematic pharmacology database and analysis platform(TCMSP)was used to screen the compound components with good activity and corresponding target proteins of Xiaoyao pill.Gene names of the target proteins were searched through Universal Protein database,and the biological species was selected as homo sapiens.The relevant gene targets were searched in Online Mendelian Inheritance in Man and Genecards databases.Protein-protein interaction network(PPI)was constructed by using protein interaction platform(STRING)to screen important targets.R language cluster Profiler software package was used to analyze GO and KEGG functional enrichment.Results Xiaoyao Pills,psychological stress-induced infertility and immunoregulation had 53 common targets,and PPI network topology analysis showed 50 related targets such as interleukin(IL)-6,signal transduction and transcriptional activation factor 3(STAT3),tumor necrosis factor(TNF),IL-1βand mitogen-activated protein kinase 1(MAPK1).Enrichment analysis showed that the signaling pathways mainly focused on IL-17 and TNF signaling pathways.Conclusion Xiaoyao Pill may immunomodulate psychological stress-induced infertility through IL-17 and TNF signaling pathways,which may provide theoretical basis for further experimental research and clinical application.
