摘要
目的:探讨环状RNA PVT1(circPVT1)在原发性非小细胞肺癌(NSCLC)患者组织和血液中的表达情况及其在NSCLC早期诊断的应用价值。方法:选取本院收治的68例NSCLC患者作为病例组,另选取同期于本院进行体检的68例正常人作为对照组。采用实时荧光定量PCR(RT-qPCR)检测病例组的肺癌组织与癌旁正常组织及两组血清circPVT1表达情况,分析circPVT1表达水平及其与NSCLC临床病理特征的关系,应用ROC曲线分析circPVT1对于NSCLC早期诊断的应用价值。结果:病例组肺癌组织circPVT1表达水平高于癌旁正常组织(P<0.05)。病例组血清circPVT1表达水平高于对照组(P<0.05)。不同性别、年龄、病理类型、肿瘤大小、组织学分化程度的肺癌组织circPVT1表达水平比较,差异均无统计学意义(P>0.05),是否吸烟、不同肿瘤分期、有无淋巴结转移的肺癌组织circPVT1表达水平比较,差异均有统计学意义(P<0.05)。组织circPVT1表达水平诊断NSCLC的ROC曲线下面积为0.805[P<0.001,95%CI(0.731,0.880)]。血清circPVT1表达水平诊断NSCLC的ROC曲线下面积为0.782[P<0.001,95%CI(0.704,0.860)]。结论:circPVT1在NSCLC组织和血清中的表达水平均明显增高,肿瘤组织中circPVT1高表达与吸烟、肿瘤分期及淋巴结转移有关,circPVT1对于NSCLC具有一定的诊断价值。
Objective:To investigate the expression of circular RNA PVT1(circPVT1)in tissues and blood of patients with primary non-small cell lung cancer(NSCLC)and its application value in early diagnosis of NSCLC.Method:A total of 68 NSCLC patients admitted to our hospital were selected as the case group,and 68 normal people who underwent physical examination in our hospital during the same period were selected as the control group.Real-time fluorescence quantitative PCR(RT-QPCR)was used to detect the expression of circPVT1 in lung cancer tissues and adjacent normal tissues as well as the serum of the two groups,the expression level of circPVT1 and its relationship with the clinicopathological features of NSCLC were analyzed,the application value of circPVT1 for the early diagnosis of NSCLC was analyzed by ROC curve.Result:The expression level of circPVT1 in lung cancer tissues was higher than that in adjacent normal tissues(P<0.05).The serum circPVT1 expression level in case group was higher than that in control group(P<0.05).There was no significant difference in the expression level of circPVT1 in lung cancer tissues of different genders,ages,pathological types,tumor sizes and histological differentiation degrees(P>0.05),but there were statistically significant differences in circPVT1 expression levels in lung cancer tissues with or without smoking,different tumor stages and with or without lymph node metastasis(P<0.05).The area under ROC curve for diagnosis of tissue circPVT1 expression level in NSCLC was 0.805[P<0.001,95%CI(0.731,0.880)].The area under ROC curve of serum circPVT1 expression level for diagnosis of NSCLC was 0.782[P<0.001,95%CI(0.704,0.860)].Conclusion:The expression level of circPVT1 in NSCLC tissues and serum was significantly increased,the high expression of circPVT1 is associated with smoking,tumor stage and lymph node metastasis,circPVT1 has a certain diagnostic value for NSCLC.
