PTEN基因突变、mRNA表达与卵巢癌病理生物学特征的相关性探讨 被引量:1

Correlation between PTEN Gene Mutation,mRNA Expression and Pathological and Biological Characteristics of Ovarian Cancer

摘要 目的:探讨第10号染色体缺失的磷酸酶和张力蛋白同源物基因(PTEN)基因突变、mRNA表达与卵巢癌病理生物学特征的相关性。方法:选择2018年1月-2020年2月本院62例卵巢癌患者作为观察组,另选择同期于本院治疗的48例卵巢良性肿瘤患者作为对照组。对照组术中取病灶组织,观察组术中取卵巢癌组织与癌旁组织。采用PCR-SSCP完成观察组基因突变检测;采用实时荧光PCR法测定不同组织中PTEN mRNA水平;分析PTEN mRNA水平与卵巢癌病理生物学特征的相关性。结果:卵巢癌组织PTEN基因突变分析结果显示,外显子5第5位密码子T突变为C,经Blast P比较发现氨基酸由丝氨酸(S)变为苯丙氨酸(F)。观察组中卵巢癌组织PTEN mRNA水平低于癌旁组织和对照组中浆液性囊腺瘤与黏液性囊腺瘤组织(P<0.05)。不同年龄、组织学类型及组织分级的卵巢癌患者PTEN mRNA水平比较,差异均无统计学意义(P>0.05);不同FIGO分期、有无淋巴结转移的卵巢癌患者PTEN mRNA水平比较,差异均有统计学意义(P<0.05)。PTEN mRNA水平与FIGO分期、淋巴结转移均呈负相关(P<0.05)。结论:PTEN基因突变参与卵巢癌的发生、发展,且PTEN mRNA在癌组织呈低表达,与卵巢癌FIGO分期、淋巴结转移存在相关性。 Objective:To investigate the correlation between the mutation and mRNA expression of the phosphatase and tensin homolog gene(PTEN)gene deletion on chromosome 10 and the pathobiological characteristics of ovarian cancer.Method:From January 2018 to February 2020,62 patients with ovarian cancer in our hospital were selected as the observation group,another 48 patients with benign ovarian tumors treated during the same period in our hospital were selected as the control group.The focal tissue was taken from the control group,and the ovarian cancer tissue and paracancer tissue were taken from the observation group.PCR-SSCP was used to detect gene mutation in the observation group.PTEN mRNA levels in different tissues were determined by real-time fluorescence PCR.The correlation between PTEN mRNA level and the pathologic characteristics of ovarian cancer was analyzed.Result:Analysis of PTEN mutations in ovarian cancer tissue showed that,codon T at position 5 of exon 5 mutation to C,the amino acid changed from serine(S)to phenylalanine(F)by comparison with Blast P.The PTEN mRNA level in the observation group was lower than those in the paracancer tissues and serous cystadenoma and myxoid cystadenoma tissues in the control group(P<0.05).There was no significant difference in PTEN mRNA level among different age,histological type and tissue grade of ovarian cancer patients(P>0.05).PTEN mRNA levels were statistically significant in ovarian cancer patients with different FIGO stages and with or without lymph node metastasis(P<0.05).PTEN mRNA level was negatively correlated with FIGO stage and lymph node metastasis(P<0.05).Conclusion:PTEN gene mutation is involved in the occurrence and development of ovarian cancer,and PTEN mRNA expression is low in cancer tissues,which is correlated with ovarian cancer FIGO stage and lymph node metastasis.
