摘要
PCNP是一种新近发现的含有PEST序列的核蛋白,主要定位于细胞核中,在体内外均可为泛素连接酶NIRF(Np95/ICBP90-like RING figer protein)作用而泛素化降解。PEST结构域是一种富含脯氨酸(Proline,P)、谷氨酸(Glutamic acid,E)、丝氨酸(Serine,S)和苏氨酸(Threonine,T)的肽序列,对于细胞周期和增殖具有调控作用,并且和染色体的稳定、肿瘤的发生以及免疫系统有关。人类恶性肿瘤预后较差,患者生存质量每况愈下,发病机制复杂多变,有研究发现PCNP的表达与PI3K/AKT/mTOR、JNK和Wnt等肿瘤发病重要信号通路有关,并在一定程度上影响某些免疫细胞、细胞因子的功能和效应分子的表达,触发相关免疫反应,在肿瘤的发病机制中发挥重要作用。主要就PCNP在细胞周期调控、细胞信号转导和肿瘤免疫方面进行综述。
PCNP is a newly discovered nuclear protein containing PEST sequence.It is mainly located in the nucleus.It can be degraded by the action of the ubiquitin ligase NIRF(Np95/ICBP90-like RING figer protein)both in vivo and in vitro.The PEST domain is a peptide sequence rich in Proline(P),Glutamic acid(E),Serine(S)and Threonine(T).It has a regulatory effect on cell cycle and proliferation.Itis alsorelated to the stability of chromosomes,the occurrence of tumors and the immune system.The prognosis of human malignant tumors is poor and the quality of life in patients is deteriorating,and the pathogenesis is complex and changeable.Studies have found that the expression of PCNP is related to important signal pathways such as PI3K/AKT/mTOR,JNK and Wnt,and affects the function of certain immune cells,cytokines and the expression of effector molecules to a certain extent,triggering related immune responses that plays an important role in the pathogenesis of tumors.Therefore,this article mainly reviews the aspects of PCNP in cell cycle regulation,cell signal transduction and tumor immunity.
