炒酸枣仁对地西泮诱导的睡眠及宿醉效应的影响

Effect of fried Semen Ziziphi spinosae on sleep and hangover induced by diazepam in mice

目的研究炒酸枣仁对地西泮所致小鼠睡眠的协同作用及对地西泮诱导的宿醉效应的影响。方法以小鼠睡眠时间和入睡率为评价指标,筛选地西泮诱导小鼠睡眠的最小有效剂量;小鼠连续7 d灌胃给予炒酸枣仁(80 g·kg^(-1)),以小鼠睡眠持续时间、睡眠潜伏期、自主活动及高架十字迷宫为评价指标,探讨炒酸枣仁对地西泮催眠的协同作用及对地西泮所致宿醉效应的影响。结果随着给药剂量的增加,地西泮诱导的小鼠睡眠时间和入睡率增加,呈现良好的量效关系,当剂量达到20.8 mg·kg^(-1)时,小鼠入睡率达到100%。与地西泮组小鼠相比,炒酸枣仁联合地西泮组小鼠的睡眠持续时间增加,自主活动次数、站立次数增加,在高架十字迷宫的开放臂进入次数及闭臂进入次数明显增多。结论炒酸枣仁对地西泮诱导的睡眠具有协同作用,对地西泮所致的宿醉效应具有拮抗作用。

Objective To determine the effect of fried Semen Ziziphi spinosae on diazepam-induced sleep and hangover effect in mice.Methods The sleep time and falling asleep rate were used as indicators to evaluate the minimum effective dose of diazepam.Then the mice were given fried Semen Ziziphi spinosae(80 g·kg^(-1))via gavage for 7 consecutive days.The sleep duration,sleep latency,locomotion behavior test and the elevated plus maze test of mice were used to determine the synergistic effect of fried Semen Ziziphi spinosae on the hypnotic and the hangover effect induced by diazepam in mice.Results As the administration dose of diazepam increased,the sleep time and falling asleep rate increased,showing a dose-dependance.When the dose of diazepam reached 20.8 mg·kg^(-1),the falling asleep rate of mice reached 100%.Compared with that of the diazepam group,the sleep duration of the fried Semen Ziziphi spinosae combining diazepam group was increased,the number of autonomous activities and standing times were significantly increased,and the number of open arm and close arm entries were significantly increased.Conclusion Fried Semen Ziziphi spinosae has a synergistic effect on the sleep induced by diazepam and an antagonistic effect on the hangover caused by diazepam.
