不同产地肉苁蓉饮片HPLC指纹图谱与化学模式识别 被引量:2

HPLC Fingerprint and Chemical Pattern Recognition of Cistanches Herba Decoction Pieces from Different Origins

摘要 目的:建立肉苁蓉饮片的高效液相色谱法(HPLC)指纹图谱,结合化学模式识别技术,为不同产地肉苁蓉饮片质量评价提供参考。方法:收集10批不同产地肉苁蓉样品。色谱柱为AgilentZORBAXC18;流动相为乙腈-0.5%乙酸(梯度洗脱);流速为1.0 mL·min^(-1);检测波长为330 nm;柱温为30℃。采用“中药指纹图谱相似度评价系统”2012 A版计算相似度,结合聚类分析和主成分分析对肉苁蓉饮片进行模式识别。结果:建立了肉苁蓉饮片的指纹图谱,确定了17个共有峰,并通过对照品比对指认了其中8个峰。不同产地肉苁蓉饮片相似度>0.94。10批肉苁蓉样品可聚为2类,分析确定了4个主成分。结论:建立的HPLC指纹图谱方法准确高效、灵敏度高,可为不同产地肉苁蓉饮片质量控制和评价提供参考。 Objective:To combine high performance liquid chromatography(HPLC)fingerprint with chemical pattern recognition for quality evaluation of Cistanches Herba decoction pieces and thereby lay a basis for the identification and quality evaluation of Cistanches Herba decoction pieces of different origins.Methods:A total of 10 batches Cistanches Herba decoction pieces of different origins were collected.HPLC conditions are as follows:Agilent ZORBAX C18 column,gradient elution with mobile phase of acetonitrile-0.5%acetic acid solution at the flow rate of 1.0 mL·min^(−1),detection wavelength of 330 nm,column temperature of 30℃.The similarity was calculated with the similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine(version 2012A).Cluster analysis and principal component analysis were employed for pattern recognition of Cistanches Herba decoction pieces.Results:The HPLC fingerprint of Cistanches Herba decoction pieces was established.A total of 17 peaks were recognized as common peaks in the fingerprint and 8 peaks were identified by comparison with the reference substances.The 10 batches of samples demonstrated the similarity of>0.94.They clustered into 2 clades and 4 principal components were identified.Conclusion:The combination of HPLC fingerprint with chemical pattern recognition is accurate,efficient,and sensitive,which can be used for quality control and quality evaluation of Cistanches Herba decoction pieces of different origins.
