摘要
目的优选关黄柏药材中生物碱的提取工艺。方法以关黄柏药材中5个生物碱总含量为评价指标,通过单因素试验结合Box-Behnken响应面法,研究甲醇体积分数、液料比和提取时间对关黄柏生物碱总含量的影响,优选最佳提取工艺,并进行验证。结果最佳提取工艺为甲醇体积分数为68%,液料比为62 m L/g,提取时间为41 min。验证试验中,5个生物碱总含量平均值为3.005 mg/g,与模型预测值的相对误差为0.59%,在允许的误差范围内。结论优选的工艺操作简便、效率高、稳定可靠,适用于关黄柏药材中生物碱类成分的提取。
Objective To optimize the extraction process of alkaloids from Phellodendri Amurensis Cortex.Methods Taking the total content of five alkaloids in Phellodendri Amurensis Cortex as the evaluation index,the effects of methanol volume fraction,liquid-material ratio and extraction time on the total content of alkaloids in Phellodendri Amurensis Cortex were studied by the single factor test and Box-Behnken response surface methodology,and the extraction process was optimized and verified.Results The optimal extraction process was as follows:the volume fraction of methanol was 68%,the liquid-material ratio was 62 m L/g,and the extraction time was 41 min.In the validation test,the average total content of five alkaloids was 3.005 mg/g,and the relative error was 0.59%compared with the predicted value of the model,which was within the allowable error range.Conclusion The optimal process is simple,efficient,stable and reliable,which is suitable for the extraction of alkaloids from Phellodendri Amurensis Cortex.
