酶法水解猴头菇多肽的生物活性 被引量:11

Bioactivities of Peptides Derived from Hericium erinaceus Proteins by Enzymatic Hydrolysis

摘要 本实验用木瓜蛋白酶、风味蛋白酶、碱性蛋白酶、胰蛋白酶对猴头菇蛋白进行酶解,得到4种猴头菇多肽HEPH-P、HEPH-F、HEPH-A、HEPH-T,通过比较不同猴头菇多肽的抗氧化活性、抗炎活性、抗肿瘤活性,研究酶法水解猴头菇多肽的生物活性。结果表明,HEPH-P具有良好的1,1-二苯基-2-三硝基苯肼自由基和羟自由基清除活性,清除率分别达到(53.01±0.27)%和(55.73±1.52)%。HEPH-A在脂多糖诱导RAW264.7细胞的抗炎模型中通过有效控制NO和白细胞介素(interleukin,IL)-6的释放、促进IL-10的分泌,从而表现出抗炎活性。HEPH-P使肝癌HepG-2细胞细胞膜破裂并且触发细胞凋亡,从而有效地抑制肝癌细胞增殖。该研究可为猴头菇功能活性肽的制备及开发应用提供理论依据。 In this study,papain,flavourzyme,alcalase and trypsin were separately used to hydrolyze Hericium erinaceus proteins,yielding peptides designated as HEPH-P,HEPH-F,HEPH-A,and HEPH-T,respectively.Their antioxidant,antiinflammatory activity,and antitumor activities were evaluated.The results showed that HEPH-P had good 2,2-di(4-tertoctylphenyl)-1-picrylhydrazyl(DPPH)radical and hydroxyl radical-scavenging activities,with scavenging percentages of(53.01±0.27)%and(55.73±1.52)%,respectively.HEPH-A showed anti-inflammatory activity by effectively controlling the release of NO and interleukin(IL)-6 and promoting the secretion of IL-10 in lipopolysaccharide(LPS)-induced RAW264.7 cells.HEPH-P resulted in the rupturing of the cell membrane,triggered apoptosis and ultimately inhibited the proliferation of liver cancer cells.This study can provide a theoretical basis for the development and application of functionally active peptides from Hericium erinaceus proteins.
