摘要
目的研究第10号染色体缺失的磷酸酶和张力蛋白同源基因(PTEN)、基质金属蛋白酶-2(MMP-2)、蛋白激酶B(AKT)在乳腺癌组织中的表达及相关性。方法收集2014年1月至2015年12月乳腺癌患者(76例)、乳腺良性病变(31例)组织标本,采用免疫组化方法检测PTEN、MMP-2、AKT阳性表达,分析PTEN阳性表达与MMP-2、AKT的相关性。结果乳腺良性病变组PTEN阳性表达高于乳腺癌组,MMP-2、AKT阳性表达低于乳腺癌组(P<0.05)。高组织学分级、肿瘤直径≥3 cm、有淋巴结转移、生存期<5年乳腺癌组织,PTEN阳性表达低于低组织学分级、肿瘤直径<3 cm、无淋巴结转移、生存期≥5年组(P<0.05),MMP-2、AKT阳性表达高于低组织学分级、肿瘤直径<3 cm、无淋巴结转移、生存期≥5年组(P<0.05)。PTEN表达与MMP-2、AKT表达呈负相关性(r=-0.634,-0.512,P<0.05)。结论PTEN表达缺失通过负向调PI3K/AKT信号通路,诱导AKT、MMP-2过度表达,共同参与乳腺癌发生发展过程。
Objective To investigate the expressions and correlation of PTEN,MMP-2 and AKT in breast cancer tissues.Methods A total of 95 tissue samples of 76 patients with breast cancer and 31 patients with benign breast lesions who were treated in our hospital from January 2014 to december 2015 were enrolled in the study.The expression levels of PTEN,MMP-2 and AKT were detected by immunohistochemistry,moreover the correlation between the positive expression of PTEN and MMP-2 as well as AKT was analyzed.Results The expression levels of PTEN in benign breast lesions group were significantly higher than those in breast cancer group,however the expression levels of MMP-2 and AKT were significantly lower than those in breast cancer group(P<0.05).The expression levels of PTEN in breast cancer tissues with high histological grade,tumor diameter≥3cm,lymph node metastasis,survival time<5 years,were significantly lower than those of breast cancer tissues with low histological grade,tumor diameter<3cm,no lymph node metastasis,survival tiem≥5 years(P<0.05).The expression levels of PTEN were negatively correlated with those of MMP 2 and AKT(P<0.05).Conclusion The expression deletion of PTEN can negatively regulate the PI3K/AKT signaling pathway,induce the over-expression of AKT and MMP-2,which may participate in the pathogenesis and development of breast cancer.
