摘要
目的探讨补阳还五汤对局灶性脑缺血大鼠血液流变学及NOD样受体蛋白3(NLRP3)/天冬氨酸特异性半胱氨酸蛋白酶1(Caspase-1)炎性反应的影响。方法将大鼠随机分为假手术组、模型组、补阳还五汤组、米诺环素组,每组25只,采用大脑中动脉栓塞法(MCAO)建立永久性局灶性脑缺血模型,于手术后1、3、7、14、28d处死,给予不同处理,取动脉血进行血液流变学检测,免疫组化法检测脑组织中NLRP3、Caspase-1阳性表达,ELISA法检测血清白细胞介素-1β(IL-1β)、白细胞介素-18(IL-18)含量。结果模型组各时间点与假手术组比较,血液流变学全血黏度及红细胞聚集指数,脑组织中NLRP3、Caspase-1表达以及血清IL-1β、IL-18含量升高明显(P<0.05);与模型组比较,给药3d后各用药组上述指标不同程度改善(P<0.05)。结论补阳还五汤可改善MCAO模型大鼠血液流变学,抑制NLRP3、Caspase-1的表达,从而抑制炎症反应,发挥脑保护及神经功能恢复的作用。
Objective:To investigate the effect of Buyang Huanwu Decoctionon hemorheologyand the expression of NLRP3 and Caspase-1 in inflammatory reaction of rats after focal cerebral ischemia.Methods:The rats were randomly divided into sham operation group,model group,BYHWD group and minocyline group,25 rats in eachgroup.The method of middle cerebral artery occlusion(MCAO)was used to establish a permanent focal cerebral ischemia model.After surgery of 1,3,7,14,28 d,the rats were received different process after executed.Arterial blood was used for hemorheology detection to detect the expression of NLRP3 and Caspase-1 content changes in rats brain by immune histochemical method,and the containing of IL-1β,IL-18 in rats serum was detected by enzyme-linked immunosorbent assay(ELISA).Results:Compared with the sham operation group at each time point,the blood rheology of whole blood viscosity and red blood cell aggregation index,the expression of NLRP3 and Caspase-1 in brain tissue,and the serum IL-1β and IL-18 levels in the model group increased significantly(P<0.05),compared with the model group,the above indicators of each medication group improved to varying degrees after 3 d of administration(P<0.05).Conclusion:Buyang Huanwu Decoctionon can improve the hemorheology of MCAO model rats,inhibit the expression of NLRP3 and Caspase-1,thereby inhibiting inflammation,and playing the role of brain protection and nerve function recovery.
