腰痹舒胶囊对寒湿痹阻腰痛模型大鼠血液流变学等相关指标的影响

Effect of Yaobishu Capsules on Hemorheology and Related Indexes in Rats with Cold-Dampness Blockage of Lumbar Pain

目的观察腰痹舒胶囊对小鼠的镇痛作用及对寒湿痹阻腰痛模型大鼠组织病理学、血液流变学的影响。方法将50只C57小鼠分为模型组,阳性药组及腰痹舒胶囊高、中、低剂量组,分别灌胃给予相应药物,末次给药后1h后,各组小鼠均腹腔注射0.8%冰醋酸0.2m L,记录注射后20min内小鼠出现扭体反应的次数及潜伏期。SD大鼠分为空白对照组、模型组、阳性药组及腰痹舒胶囊高、中、低剂量组,寒湿痹阻模型造模成功后各组灌胃给予相应药物,每日1次,连续7d;末次给药1h后,全部处死,测定血液流变学指标同时取各组腰部肌肉组织送病检。结果中、高剂量腰痹舒胶囊都可以延长冰醋酸所致小鼠扭体反应潜伏期及减少小鼠扭体反应次数(P<0.01),低剂量可以延长冰醋酸所致小鼠扭体反应潜伏期(P<0.05);高、中剂量腰痹舒胶囊能降低寒湿痹阻模型大鼠的低切全血黏度和低切还原黏度(P<0.05或P<0.01)。组织病理学提示腰痹舒胶囊各组与模型组自然愈合无明显差异。结论腰痹舒胶囊对小鼠有一定的镇痛作用,对寒湿痹阻模型大鼠有一定活血化瘀作用。

Objective:To observe the analgesic effect of Yaobishu Capsules on mice and the effect of histopathology and hemorheology on rats with cold-dampness blockage of lumbago.Methods:A total of 50 C57 mice were divided into the model group,positive drug group,and Yaobishu Capsules high-dose,medium-dose and low-dose groups.Relevant drugs were administered orally,respectively.One hour after the last administration,0.8%glacial acetic acid 0.2 mL was intraperitoneally injected into each group,and the times and latency of writhing reaction in mice were recorded within 20 min.SD rats were divided into blank control group,model group,positive drug group and Yaobishu Capsules high-dose,medium-dose and low-dose groups.After the model was successfully established,the remaining groups were given corresponding drugs by intragastric administration,once a day,for consecutive 7 d.One hour after the last administration,all groups were sacrificed,and the hemorheology indexes were determined.Meanwhile,the lumbar muscle tissues of each group were sent for disease examination.Results:Both medium and high doses of Yaobishu Capsules could prolong the latency of writhing reaction and reduce the number of writhing reactions in mice induced by glacial acetic acid(P<0.01),while low dose could prolong the latency of writhing reaction in mice induced by glacial acetic acid(P<0.05).High and medium doses of Yaobishu Capsules could reduce the low cut whole blood viscosity and low cut reduced viscosity of rats with cold-dampness blockage syndrome(P<0.05 or P<0.01).Histopathological findings showed that there was no significant difference in natural healing between Yaobishu Capsules groups and model group.Conclusion:Yaobishu Capsules has analgesic effect on model mice and can promote blood circulation and dissipate stasis in model rats.
