摘要
目的分析红花注射液对柯萨奇B3病毒(CVB3)诱导病毒性心肌炎(VMC)大鼠心肌组织B细胞淋巴瘤/白血病-2(Bcl-2)和Bcl相关蛋白(Bax)表达的影响。方法将50只大鼠随机分为对照组、CVB3组和红花注射液低、中、高剂量组各10只。干预后,评估心脏指数和心肌组织病理积分,苏木精伊红(HE)染色和原位末端标记法(TUNEL)检测心肌组织损伤,检测血清肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)、心肌肌钙蛋白I(cTnI)、白细胞介素(IL)-6、IL-10和肿瘤坏死因子-α(TNF-α)水平,心肌组织CVB3 mRNA、Bcl-2、Bax、裂解的半胱氨酸蛋白酶-3(cleaved Caspase-3)、Toll-样受体4(TLR4)、核因子-кB(NF-кB)p65、磷酸化NF-кB抑制蛋白(p-IκB)表达。结果CVB3组大鼠心脏指数和心肌组织细胞凋亡率高于对照组,血清CK-MB、LDH、cTnI、IL-6、TNF-α、IL-10水平高于对照组,心肌组织TLR4、NF-кB p65、p-IκB和cleaved Caspase-3蛋白表达高于对照组,Bcl-2/Bax低于对照组(P<0.05);红花注射液高、中剂量组心脏指数、心肌组织病理积分、心肌组织细胞凋亡率和CVB3 mRNA低于CVB3组,血清CK-MB、LDH、cTnI、IL-6、TNF-α水平低于CVB3组,IL-10水平高于CVB3组,心肌组织TLR4、NF-кB p65、p-IκB和cleaved Caspase-3蛋白表达低于CVB3组,Bcl-2/Bax高于CVB3组(P<0.05)。结论红花注射液可以抑制VMC大鼠炎症反应,促进Bcl-2/Bax表达,减轻心肌组织损伤。
OBJECTIVE To observe the effect of safflower injection on expression of B-cell lymphoma/leukemia-2(Bcl-2)and Bcl-associated proteins(Bax)in myocardial tissues of rats with Coxsackie virus B3(CVB3)-induced viral myocarditis(VMC).METHODS A total of 50 rats were randomly divided into the control group,the CVB3 group,the low-dose safflower injection group,the medium-dose safflower injection group and the high-dose safflower injection group,with 10 rats in each group.The cardiac index and pathological scores of myocardial tissues were evaluated,the damage of myocardial tissues was detected by HE staining and TUNEL staining.The levels of serum creatine kinase isoenzyme(CK-MB),lactate dehydrogenase(LDH),cardiac troponin I(cTnI),interleukin(IL)-6,IL-10 and tumor necrosis factor-alpha(TNF-α)were detected,and the expression levels of CVB3 mRNA,Bcl-2,Bax,cleaved Caspase-3(cleaved Caspase-3),Toll-like receptor 4(TLR4),nuclear factorкB(NF-кB)p65,phosphorylated NF-кB inhibitory protein(p-IκB)in myocardial tissues were detected.RESULTS The cardiac index and apoptosis rate of myocardial cells of the CVB3 group were signigifcantly higher than those of the control group,the levels of serum CK-MB,LDH,cTnI,IL-6,TNF-αand IL-10 of the CVB3 group were signigifcantly higher than those of the control group,the expression levels of TLR4,NF-кB p65,p-IκB and cleaved Caspase-3 proteins in myocardial tissues were signigifcantly higher in the CVB3 group than in the control group,and the Bcl-2/Bax of the CVB3 group was lower than that of the control group(P<0.05).The cardiac index,pathological scores of myocardial tissues,apoptosis rate of myocardial cells and CVB3 mRNA of the medium-dose safflower injection group and the high-dose safflower injection group were signigifcantly lower than those of the CVB3 group,the levels of serum CK-MB,LDH,cTnI,IL-6 and TNF-αof the medium-dose safflower injection group and the high-dose safflower injection group were signigifcantly lower than those of the CVB3 group,the IL-10 level of the medium-dose safflower injection group and the high-dose safflower injection group was signigifcantly higher than that of the CVB3 group,the expression levels of TLR4,NF-кB p65,p-IκB and cleaved Caspase-3 in myocardial tissues were signigifcantly lower in the medium-dose safflower injection group and the high-dose safflower injection group than in the CVB3 group,and the Bcl-2/Bax of the medium-dose safflower injection group and the high-dose safflower injection group was signigifcantly higher than that of the CVB3 group(P<0.05).CONCLUSION The safflower injection can inhibit the inflammatory response in the VMC rats,promote the expression of Bcl-2/Bax and alleviate the damage of myocardial tissues.
