胰腺癌组织中SOX4和E-cadherin的表达及临床意义 被引量:9

Expression of SOX4 and E-cadherin in pancreatic carcinoma and their clinical significance

摘要 目的探讨胰腺癌组织中上皮-间质转化(epithelial-mesenchymal transition,EMT)相关分子SOX4和E-cadherin蛋白的表达及临床意义。方法采用免疫组化SP法检测125例胰腺癌组织及其对应癌旁组织中SOX4和E-cadherin的表达,分析两者表达与胰腺癌临床病理特征及预后的关系。结果胰腺癌组织中SOX4蛋白阳性率高于相应癌旁组织(77.6%vs 12.0%,P<0.05)。SOX4蛋白表达与胰腺癌肿瘤大小、肿瘤部位、AJCC分期、淋巴结转移、脉管内癌栓和神经侵犯相关(P<0.05)。胰腺癌组织中E-cadherin蛋白阳性率显著低于相应癌旁组织(14.4%vs 82.4%,P<0.05)。E-cadherin蛋白表达与胰腺癌肿瘤大小、肿瘤部位、肿瘤分化程度、AJCC分期、淋巴结转移、脉管内癌栓和神经侵犯相关(P<0.05)。Spearman秩相关检验显示,胰腺癌组织中SOX4和E-cadherin蛋白表达呈负相关(r=-0.763,P<0.001)。Kaplan-Meier生存曲线分析显示,SOX4蛋白阳性或E-cadherin蛋白阴性患者的生存期相对缩短(P<0.001)。Cox风险模型分析显示,肿瘤大小、肿瘤分化程度、AJCC分期、淋巴结转移、脉管内癌栓、神经侵犯、SOX4蛋白表达、E-cadherin蛋白表达与患者生存期有关(P<0.05)。结论胰腺癌中SOX4蛋白表达升高和E-cadherin蛋白表达下降可能参与胰腺癌的发生、发展,联合检测SOX4和E-cadherin蛋白表达有助于胰腺癌的预后判断。 Purpose To investigate the expression and clinical significance of EMT-related molecules SOX4 and E-cadherin in pancreatic cancer tissues.Methods Immunohistochemical SP method was used to observe the expression of SOX4 and E-cadherin in 125 pancreatic cancer tissues and their adjacent tissues.The relationship was analyzed between their expression and the clinicopathological features and prognosis of pancreatic cancer.Results The positive rate of SOX4 protein in pancreatic cancer tissues was higher than that in adjacent tissues(77.6%vs 12.0%,P<0.05).The expression of SOX4 protein was significantly correlated with tumor size,tumor location,AJCC stage,lymph node metastasis,vascular cancer embolusand nerve invasion in pancreatic cancer patients(P<0.05).The positive rate of E-cadherin protein in pancreatic cancer tissues was significantly lower than that in adjacent tissues(14.4%vs 82.4%,P<0.05).The expression of E-cadherin protein was correlated with tumor size,tumor location,tumor differentiation degree,AJCC stage,lymph node metastasis,vascular cancer embolus and nerve invasion in pancreatic cancer patients(P<0.05).Spearman rank correlation test showed a negative correlation between SOX4 and E-cadherin protein expression in pancreatic cancer(r=-0.763,P<0.001).Kaplan-Meier survival analysis showed that patients with positive SOX4 expression or negative E-cadherin expression had a relatively shorter survival time(P<0.001).Cox risk model analysis showed that tumor size,differentiation degree,AJCC stage,lymph node metastasis,vascular cancer embolus,nerve invasion,SOX4 protein expression and E-cadherin protein expression were correlated with survival time(P<0.05).Conclusion The increased expression of SOX4 protein and the decreased expression of E-cadherin protein in pancreatic cancer may be involved in the occurrence and development of pancreatic cancer.Combined detection of SOX4 and E-cadherin protein expression can help predict the prognosis of pancreatic cancer.
