基于感官特征结合灰色关联度法评价积雪草质量 被引量:10

Quality evaluation of Centella asiatica based on sensory characteristics and gray relational analysis

摘要 目的:结合感官特征与灰色关联度法对积雪草质量进行综合评价,以提高积雪草质量评价的可靠性及科学性。方法:采用生药学方法对不同产地积雪草进行性状、显微结构及薄层色谱等鉴别;参考《中华人民共和国药典》四部通则测定其杂质、水分、总灰分和浸出物含量;采用HPLC法对积雪草苷、羟基积雪草苷进行含量测定;采用灰色关联度法构建积雪草质量评价模型,计算其相对关联度。结果:不同产地积雪草的药材性状、显微结构差异不显著;薄层色谱均能检测出积雪草苷和羟基积雪草苷;杂质含量为0.500%~8.837%;水分含量为4.899%~6.423%;总灰分含量为10.103%~16.368%;浸出物含量为28.100%~38.311%;积雪草苷含量为0.383%~1.019%;羟基积雪草苷含量为0.541%~1.915%;综合杂质、水分、总灰分、浸出物、积雪草苷和羟基积雪草苷等因素,不同产地的29批积雪草存在质量差异,其相对关联度值的范围为0.437~0.556,排名前3的样品为S15,S8,S9,其中江西省吉安市峡江县产积雪草,测得的相对关联度较大,质量排序靠前,表明江西省吉安市峡江县产积雪草质量较佳。结论:通过感官特征结合灰色关联度法所建立的质量评价模型可用于不同产地积雪草的质量评价。 Objective:To comprehensively evaluate the quality of Centella asiatica by combining the sensory characteristics and gray relational analysis to improve the reliability and scientificity of the quality evaluation of Centella asiatica.Methods:The characteristics,microscopic structure and thin layer chromatography of Centella asiatica were identified by pharmacognosy methods;the impurity,moisture,total ash and extract content were determined with reference to the fourth general rule of the Chinese Pharmacopoeia;HPLC method was used to determine the contents of impurities,moisture,total ash and extracts of Centella asiatica.The contents of asiaticoside and madecassoside were determined;the gray relational analysis was used to construct the quality evaluation model of Centella asiatica,and the relative degree of correlation was calculated.Results:There were no significant differences in the characteristics and microscopic structure of Centella asiatica from different producing areas;thin layer chromatography could detect both asiaticoside and madecassoside;the impurity content was 0.500%~8.837%;the moisture content was 4.899%~6.423%;the total ash content was 10.103%~16.368%;the extract content was 28.100%~38.311%;the asiaticoside content was 0.383%~1.019%;the madecassoside content was 0.541%~1.915%.Combined with various factors such as impurities,moisture,total ash,extract,asiaticoside and madecassoside,it was indicated that the 29 batches of Centella asiatica from different producing areas had quality differences and their relative correlations degree values were in the range of 0.437~0.556.The top 3 samples are S15,S8,and S9,Centella asiatica produced in Xiajiang county,Ji’an city,Jiangxi province,the measured relative degree of correlation is larger,and the quality ranking is higher,indicating that the quality of Centella asiatica produced in Xiajiang county,Ji’an city,Jiangxi province is better.Conclusion:The quality evaluation model established by the sensory characteristics combined with gray relational analysis can be used for the quality evaluation of Centella asiatica from different producing areas.
