基于指纹图谱结合化学计量法对不同干燥方式下西洋参皂苷类成分的分析 被引量:13

Analysis of Saponins of Xiyangshen(American Ginseng) under Different Drying Methods Based on Fingerprint and Chemometric Method

摘要 采用HPLC法对12批不同批次西洋参饮片6种皂苷类成分进行测定,运用国家药典委员会"中药色谱指纹图谱相似度评价软件(2012版)"进行评价,并结合聚类分析和主成分分析等化学计量法对4种不同干燥方式下(普通晾干组、60℃初始烘干组、30℃初始烘干组、冻干组)的西洋参饮片进行比较和区分。建立了西洋参皂苷类成分HPLC指纹图谱,相似度均达0.940以上,指认了6个共有峰(Rg1、Re、Rb1、Rc、Rb2、Rd)构成西洋参饮片的特征峰。聚类分析和主成分分析结果相似,30℃初始烘干组、冻干组比较明显的分为两类,但是60℃初始烘干组和普通晾干组区分不是特别明显。该法所建立的西洋参皂苷类成分指纹图谱特征性强、方法简便,结合6种皂苷类成分含量测定可更好控制其质量,对西洋参饮片的鉴定、质量控制具有指导意义和参考价值。 Six saponins of 12 different batches of Xiyangshen(American ginseng) decoction pieces were determined by HPLC, and evaluated by Chinese Medicine Chromatographic Fingerprint Similarity Evaluation Software(2012 Version) published by National Pharmacopoeia Commission, combined with cluster analysis and principal component analysis. The metering method was used to compare and distinguish the Xiyangshen(American ginseng) decoction pieces under 4 different drying methods(common drying group, 60 ℃ initial drying group, 30 ℃ initial drying group, freeze-drying group). The HPLC fingerprint of the ginseng saponins was established, and the similarity was above 0.940. Six common peaks(Rg1, Re, Rb1, Rc, Rb2, Rd) were identified as the characteristic peaks of Xiyangshen(American ginseng) decoction. The results of cluster analysis and principal component analysis were similar. The initial drying group and the freeze-drying group at 30 ℃ were obviously divided into two categories, but the distinction between the initial drying group at 60 ℃ and the ordinary drying group was not particularly obvious. The fingerprint of Xiyangshen(American ginseng) saponins established by this method has strong characteristics and simple methods. Combined with the determination of the contents of 6 kinds of saponins, the quality can be better controlled, which has guiding significance and reference value for the identification and quality control of Xiyangshen(American ginseng) decoction pieces.
