摘要
目的用主成分分析及聚类分析建立一种中药多指标成分的质量评价方法,优选最佳葛根趁鲜切制规格,为葛根的产地加工和鲜切工艺提供参考。方法趁鲜切制15种不同规格葛根(包括丁、片、丁片3种片型),结合不同规格饮片的外观评价,对其溶出液、煎出液中葛根素、3’-羟基葛根素、3’-甲氧基葛根素、葛根素芹菜糖苷、大豆苷、芒柄花苷、大豆苷元、染料木苷化学成分定量分析,优选出葛根鲜切规格。结果三种片型下丁片、丁整体优于片型,15种不同规格饮片中以丁片(1.5 cm×0.4 cm×0.4 cm)最优,丁(0.6 cm×0.6 cm×0.6 cm)其次。结论优选出最佳规格葛根饮片的方法具有全面性、实用性,所得参数可为葛根产地加工提供理论依据,为葛根的研究生产提供科学依据,为规格标准的制定提供一定参考价值。
Objective To establish a quality evaluation method of multi-target ingredients of traditional Chinese medicine by principal component analysis and cluster analysis, and optimize the best specification of fresh-cut Gegen(Pueraria lobata), so as to provide a reference for the processing in producing area and fresh-cut technology of Gegen(Pueraria lobata). Method A total of 15 fresh-cut Gegen(Pueraria lobata) decoction pieces(including 3 tablet-types, namely dice, slice and dice and slice) with different specifications were prepared. Combined with the appearance evaluation of 15 fresh-cut Gegen(Pueraria lobata) decoction pieces with different specifications and the contents of puerarin, 3’-hydroxy puerarin, 3’-methoxy puerarin, puerarin apioside, daidzin, ononin, daidzein, genistin and others in dissolution solution and decoction solution, the related investigations were carried out. Based on the processing experience, traditional habits and modern analysis methods of producing area, the fresh cut specifications of Gegen(Pueraria lobata) were optimized. Result Among the 3 tablet-types, dice and slice as well as dice were better than slice on the whole. Dice and slice(1.5 cm×0.4 cm×0.4 cm) was the bestin 15 decoction pieces with different specifications, followed by dice(0.6 cm×0.6 cm×0.6 cm). Conclusion The method selecting the best specification of Gegen(Pueraria lobata) decoction pieces is comprehensive and practical and the obtained parameters can provide theoretical basis for processing for the processing of Gegen(Pueraria lobata) in producing area, scientific basis for the research and production of Gegen(Pueraria lobata), and certain reference value for the formulation of specifications and standards.
