HPLC法测定川黄柏不同炮制品中盐酸小檗碱的含量 被引量:9

摘要 目的探讨中药川黄柏不同炮制品中盐酸小檗碱含量的测定结果。方法以生川黄柏、盐炙川黄柏、川黄柏炭、清炒川黄柏为对照,Agilent-C_(18)(250 mm×4.6 mm, 2.5μm)色谱柱,流动相:乙腈-0.1%磷酸缓冲溶液(55∶45),检测波长为265 nm,柱温25℃,进样量为15μL。结果 (1)盐酸小檗碱的线性回归方程为:Y=4.032 8×10^(6)X+1.998 7×10^(5)(r=0.999 8),在0.467 9~4.679μg范围内线性关系较好;(2)与生川黄柏比较,川黄柏炭中的盐酸小檗碱含量最低,清炒川黄柏含量最高,盐炙川黄柏次之;(3)随着温度的升高,不同炮制品中的盐酸小檗碱的含量会逐渐降低。结论采用高效液相色谱法可以准确测定川黄柏不同炮制品中有效成分盐酸小檗碱的含量,方法简单,易行,结果准确,同时测定出经清炒后的川黄柏中小檗碱的含量是最高的,从而为川黄柏不同炮制品在临床的应用提供参考。 Objective To investigate the determination results of berberine hydrochloride in different processed products of Chuanhuangbai(Phellodendron chinense Schneid)(PCS).Methods Taking raw PCS,salt-processed PCS,charcoal PCS and stir fried PCS were as control.The column was packed with Agilent-C_(18) column(250 mm×4.6 mm,2.5μm).The mobile phase was acetonitrile-0.1%phosphate buffer solution(55∶45),the column temperature was 25℃,the detection wavelength was 265 nm,and the injection volume was 15μL.Results①The linear regression equation of berberine hydrochloride was:Y=4.0328×10^(6) X+1.9987×10^(5)(r=0.9998),and the linear relationship was better in the range of 0.4679~4.679μg.②Compared with raw PCS,the content of berberine hydrochloride in charcoal PCS was the lowest,and the content of stir fried PCS was the highest,followed by the content in salt-processed PCS.③With the increase of temperature,the content of berberine hydrochloride in different processed products would gradually decrease.n Conclusion High performance liquid chromatography(HPLC)can accurately determine the content of effective component berberine hydrochloride in different processed products of PCS.The method is simple,easy and accurate.At the same time,the determined content of berberine in stir fried PCS is the highest,which provides a reference for the clinical application of different processed products of PCS.
