高效液相色谱测定全蝎中多西环素的含量 被引量:1

Determination of Doxycycline in Scorpio by High Performance Liquid Chromatography

摘要 目的:测定不同产地全蝎中多西环素的含量,研究该动物药材养殖过程中抗生素的使用情况。方法:使用高效液相色谱法,色谱柱为Agilent C18(柱长250mm,柱内直径4.6mm,填料颗粒直径5μm);流动相为醋酸铵溶液[0.15mol/L醋酸铵溶液-0.01mol/L乙二胺四醋酸二钠溶液-三乙胺(100:10:1),用醋酸调节pH值至8.5]-乙腈(83:17);流速1.0ml/min;检测波长为280nm;柱温40℃;进样量10μl;共测定6个产地10批样品。结果:10批样品中,来自产地A的4批多西环素含量最高,为2.111-2.364mg/g;其次为产地D的1批,含量0.873mg/g,其余B、C、E、F等地的5批无明显差异,含量在0.003-0.013mg/g之间。结论:本研究所用方法快速、准确、高效,为研究全蝎饲养中的抗生素添加提供了技术支持。 Objective: through determination of Doxycycline contents in Scorpio of different origins, to investigate antibiotic usage in raising animal TCM ingredient;Method: a High Performance Liquid Chromatography(HPLC) method was introduce for content determination, the column was Agilent C18(column length 250 mm, inner diameter 4.6 mm, packed particle diameter 5μm), with a mobile phase containing Ammonium acetate solution[0.15 mol/L Ammonium acetate-0.01 mol/L Disodium Edetate-Triethylamine(100:10:1), pH adjusted to 8.5 by acetic acid)-acetonitrile(83:17), the flowrate was 1.0 ml/min, the detection wavelength was 280 nm, the column temperature was 40℃ and the injection volume was 10μl;10 batches of samples from 6 different origins were analyzed;Result: in 10 batches of samples, 4 batches from origin A had highest Doxycycline content, which was 2.111-2.364 mg/g;the one next to it was 1 batch from origin F, the value was 0.873 mg/g;the rest 5 batches from B, C, E, F had no significant difference,and the value was 0.003-0.013 mg/g. Conclusion: the method used in this work was fast, accurate and efficient, providing technical support to the investigation of antibiotic addition in raising Scorpio.
