摘要
目的分析探讨外源性雄激素对人包皮成纤维细胞(HFF-1)基因表达的影响。方法通过培养人包皮成纤维细胞系(HFF-1),将细胞按照有无雄激素处理分为实验组和对照组,实验组用含乙醇的二氢睾酮刺激,对照组只用乙醇刺激,然后利用转录组测序技术(RNA-seq)宏观分析差异表达的基因,并进行相关生物信号通路分析,进一步通过定量PCR对部分差异表达基因进行验证。结果雄激素处理后能显著升高或抑制部分基因的表达,且主要富集于细胞外基质结构及细胞外成分、细胞迁移能力信号通路、胶原蛋白结合相关基因表达信号通路,及细胞增殖凋亡迁移的信号通路和炎症信号通路等。结论雄激素可能通过表观遗传调控的方式影响相关基因的细胞外基质结构及细胞外成分表达,进而调控HFF-1在病理疤痕纤维化过程的重要作用。
Objective To analyze the effect of exogenous androgens on gene expression of human foreskin fibroblasts(HFF-1).Methods Human foreskin fibroblasts were cultured and divided into the experimental group and the control group according to the presence or absence of androgen treatment.The cells in the experimental group were stimulated with alcohol-containing dihydrotestosterone,and the cells in the control group were only stimulated with alcohol.Their differentially expressed genes were screened by transcriptome sequencing technology(RNA-seq),and the related biological signal pathways were analyzed.Some differentially expressed genes were validated by quantitative PCR.Results Androgen intervention significantly increased or decreased the expression of some genes,and these genes were mainly enriched in the extracellular matrix structure and extracellular components,the signal pathway of cell migration,the signal pathway of collagen binding-related gene expression,and cell proliferation and apoptosis.migration signaling pathways and inflammation signaling pathways,etc.Conclusion Androgens may affect the extracellular matrix structure and the expression of extracellular components through epigenetic regulation,thereby play an important role in the pathological process of pathological scar fibrosis.
