摘要
目的:探讨采用生物标志物检测中药注射剂抑制血小板活化作用的方法适用性。方法:选择舒血宁注射液等几种具有活血化瘀作用的中药注射剂,并以无该作用的喜炎平注射液为参比。以膜糖蛋白CD62p为生物标志物,采用流式细胞术体外法检测药物对血小板活化的抑制作用。结果:舒血宁注射液、血必净注射液、注射用红花黄色素以及银杏达莫注射液可明显抑制由ADP引起的CD62p高表达,即抑制血小板的活化;而无类似作用的喜炎平注射液则为阴性结果。结论:采用流式细胞仪检测血小板活化生物标志物,能够可靠评价药物对血小板活化的抑制作用,可以作为中药注射剂活血化瘀生物活性的评价方法。
Objective:To explore the applicability of biomarkers in detecting the inhibitory effect of traditional Chinese medicine(TCM)injection on platelet activation.Methods:TCM injections with the effect of promoting blood circulation and removing blood stasis were selected,such as Shuxuening injection,Xuebijing injection,safflower yellow pigment for injection,and Ginkgo damo injection,and Xiyanping injection without the effect was used as a reference.Using membrane glycoprotein CD62p as a biomarker,the inhibitory effect of drugs on platelet activation was detected by flow cytometry in vitro.Results:Shuxuening injection,Xuebijing injection,safflower yellow pigment for injection,and Ginkgo damo injection could significantly inhibit the high expression of CD62p induced by ADP,that is,inhibit the activation of platelets.Xiyanping injection without a similar effect was negative.Conclusion:The detection of platelet activation biomarkers by flow cytometry can reliably evaluate the inhibitory effect of drugs on platelet activation and can be used as an evaluation method for the biological activity of traditional Chinese medicine injection to promote blood circulation and remove blood stasis.
